Brucellosis is an infectious disease caused by Brucella.It is widespread in the world.It not only endangers public health,but also has a significant impact on the development of animal husbandry,causing huge economic losses.Brucella mainly infects sheep,cattle and pigs,which can cause abortion of pregnant female animals and damage the reproductive system of male animals,leading to infertility.Brucella infection in humans is mainly chronic and persistent,which is characterized by wave fever,general weakness,night sweat and joint swelling and pain.Brucella is a kind of intracellular parasite.It is very important for Brucella to invade cells and survive in cells to establish persistent infection strategy.Brucella has no typical virulence system,such as exotoxin,capsule,and type III secretion system,but type IV secretion system(T4SS).T4 SS is an important virulence factor of Brucella.It can establish replication niche in endoplasmic reticulum by regulating the signal pathway of host cells,so as to inhibit host immune response and promote the formation of chronic infection.T4 SS plays a key role in mediating the survival of Brucella and inhibiting host immune response.Effector protein(also known as effector)is the substrate of type IV secretion system of Brucella.T4 SS mainly functions by secreting effector protein to act on the target of host cell.Effector proteins play an important role in the process of bacterial infection.BspE is an effector protein secreted by Brucella T4 SS.It is of great significance to study its survival and mechanism in host cells for elucidating the pathogenesis and effective prevention and control of brucellosis.In this study,bspe was constructed on p CAGGS-HA-BspE and transfected into HEK-293 T cells,and its expression was detected by WB.Then,BspE and proteins interacting with bspe were captured by IP technology,and the proteins interacting with BspE were analyzed by high-resolution bio mass spectrometry.A total of 70 host proteins were identified to be able to interact specifically with bspe,and the interaction network of the identified proteins was analyzed by software.It was found that the identified proteins were involved in the regulation of apoptosis pathway and intracellular material transport pathway,indicating that Brucella may regulate the pathway by interacting with related proteins.
