As an important behavior and cognitive process, attention is the direction and concentration of psychological activities on certain objects, that is,selecting important information and eliminating the interference of irrelevant stimuli.Attention ensures that people have a clearer understanding of things,a more accurate response, and more controlled and orderly behavior.Attention deficit behaviors include abnormalities in features such as concentration, stability, and selectivity of attention, and impulse control ability problems include patience, waiting, poor frustration tolerance, and overactivity. This thesis is based on the "top-down" information processing mode from the cortex to the striatum, focusing on the pathway between the prefrontal cortex and the caudate nucleus. The experimental object of this study is male mice, using the 5-choice serial reaction time task as the behavioral paradigm that mainly measures attention behavior level, using optogenetics and multi-channel in vivo electrophysiological techniques to explore the above problems. The findings found that: 1)During the training phase, mice meed more time to pass sessions at the stimulus presentation time of 32 seconds,2 seconds, and 1 second, and the latency period for correct and wrong responses can be gradually shortened by training.2)In the stimulus presentation stage, when the neural circuit from the mPFC to the striatum was suppressed, the accuracy of the task and the perseverative responses decreased.3)Suppression of this neural circuit during the intertrails interval will lead to an decrease in the perseverative responses after the mice responds correctly.4)Further investigation of neuron firing in the five-choice continuous response task of mice through multi-channel in vivo electrophysiological recording technology found that the frequency of firing of relevant neurons in mice at the time of touch screen increased significantly, and the moment of stimulus presentation There are fewer neurons responding. In the summary section, the experimenter analyzed and summarized the key experimental factors such as the 5-CSRTT task execution defects, the main reasons for the high omission rate in mice, and the effect of electrophysiological in vivo records and optogenetics on attention behavior to get more efficient results. The measurable attention paradigm related to the rodent model is used to specifically control the connection between the mPFC and the striatum using the neural circuit manipulation tool to block the flow of information between the circuits and investigate whether neural circuit manipulation can regulate attention performance.Further explored the neuronal firing characteristics of the anterior cingulate gyrus in regulating attention behavior. This will supply new thinking for future clinical treatment of attention-deficit-related neurological diseases.
