China is one of the countries with the richest grassland resources in the world,but the problem of grassland degradation is also very serious.Nutrient addition can directly affect soil environment and improve grassland.The research site is located in the leymus chinensis test site of Hulunbeier Grassland ecosystem national field science observation and research station of Chinese Academy of Agricultural Sciences,nutrient addition experiments were conducted under different fertilization treatments,by using high-throughput sequencing and other techniques,the change rules of soil physical and chemical properties,soil fungal community structure and diversity under different fertilization conditions were analyzed,so as to provide scientific basis for the reasonable nutrient addition and the speed of restoration of degraded grasslands.Below are key research findings:1)In this paper,seven physical and chemical properties are selected: soil total phosphorus,total carbon,total nitrogen,organic carbon,carbon-to-nitrogen ratio,carbon-to-phosphorus ratio,and nitrogen-to-phosphorus ratio.An analysis of variance was conducted in the fertilization treatment and soil depth,and it was found that there was no interaction between the indicators of total carbon,total nitrogen,organic carbon,carbon-to-nitrogen ratio,and carbon-to-phosphorus ratio,and there were interactions between the two indicators of total phosphorus and nitrogen-to-phosphorus ratio.Among them,the total phosphorus content of the CK treatment plot,N treatment plot,N1 treatment plot,N2 treatment plot,and NP treatment plot in the 5-10 cm depth soil was significantly different from that in the P treatment plot.difference.There was a significant difference between the total phosphorus content of the CK-treated plot,the N-treated plot,and the N2-treated plot in the 10-20 cm depth soil and the P plot.But there are no plots with significant differences in the depth of 20-30 cm.The ratio of nitrogen to phosphorus in the soil with a depth of 5-10 cm in the CK treatment plot,N treatment plot,N1 treatment plot,N2 treatment plot,and NP treatment plot has the same total phosphorus content as that of the P treatment plot.Significant differences.But there is no significant difference between 10-20 cm and 20-30 cm.2)A total of 9 fungal phyla were detected in the degraded leymus meadow steppe in the study area,among which Ascomycota and Basidiomycota were the dominant flora composed of various soil fungal communities.As the soil depth increased,the relative abundance of some fungi,such as Agaricales,Hypocreales,and Helotiales changed.There are a large number of unknown species to be discovered at the order,family and genus level.3)The fungal community structure of N plot and NP plot is similar.Other fertilization treatments did not cause significant differences in fungal communities.4)In this study,fertilization treatment significantly increased the diversity of soil fungi,but the increase in richness was not significant.5)In the soil surface layer(0-20cm),the high concentration of nitrogen addition will have a significant impact on the soil fungal community and change the composition of the soil fungal community.
