It is well known that lactic acid bacteria are a kind of microorganisms beneficial to human health.In this paper,we had research on the degradation of omethoate by lactic acid bacteria isolated during fermentation based on natural fermented sauerkraut.The lactic acid strains with better degradation effect were screened out and their growth characteristics and omethoate degradation characteristics were studied.Identification of the strain based on physiological and biochemical tests and 16Sr RNA gene sequence.A model of chronic organophosphorus poisoning mice was established,and different doses of Lactobacillus plantarum Z3 intervention were carried out on the mice in the model group.Through the determination of various biochemical indicators,the improvement of Lactobacillus plantarum Z3 to mice with chronic poisoning of omethoate was evaluated,and the optimal dose of administration was determined.The specific content of the study is as follows:?1?Strain Z3 with the best effect of degrading omethoate was screened from naturally fermented sauerkraut juice and was determined to be the genus Lactobacillus plantarum by the study of the colony form and the identification of 16Sr RNA.?2?The growth characteristics of degraded strains were studied,and the results showed that Lactobacillus plantarum Z3 was 0-4 h for growth delay,and 4-16 h was in the logarithmic phase and after 16h,they entered the stable phase.?3?The degradation characteristics of the strains were studied,and the results showed that the Lactobacillus plantarum Z3 could not degrade omethoate in 0-3 h,3-18 h degraded omethoate speed accelerated,and 18-24 h degradation of omethoate speed did not change significantly.?4?Determine the optimal degradation conditions of Lactobacillus plantarum Z3 for omethoate.The results of response surface analysis showed that the inoculation amount of Lactobacillus plantarum Z3 was 5%,the concentration of omethoate was 69 mg/L and the culture temperature was 41?.The degradation rate of omethoate by Lactobacillus plantarum Z3 was 23.47%.?5?Establishing a model of chronic omethoate poisoning in mice,and the detoxification effect of Lactobacillus plantarum Z3 to chronic poisoning mice of omethoate was evaluated through the intervention test of Lactobacillus plantarum Z3.The results showed that compared with the model group,the activity of cholinesterase?Ch E?in brain and whole blood of mice in high dose group increased by 0.43 U/mg prot and 22.46 U/m L;the content of malondialdehyde?MDA?in the serum and liver decreased by 0.85 nmol/m L and 0.13 nmol/mg prot;the activity of total superoxide dismutase?T-SOD?increased by 27.55 U/m L and 54.12 U/mg prot;the content of reactive oxygen species?ROS?in the brain decreased by 9.34 U/m L;and the correlation was significant?P<0.05?.Liver and brain tissue damage was reduced.The high-dose group of Lactobacillus plantarum Z3(1?1010 CFU/m L)improved the mice with chronic omethoate poisoning best.
