Behavior is an important functional manifestation of the advanced central nervous system of the organism.Whether it is studying the behavioral structure itself or studying the potential neural or genetic mechanisms of behavior,it is very important to conduct objective, quantitative and effective behavioral evaluation of organisms. In order to study the quantitative measurement of animal behavior and achieve a more objective analysis,we applied the optical motion capture system OptiTrack. In this study, the high-precision quantitative characterization method of rat behavior based on OptiTrack was studied by combining large-area open-field and optical motion capture methods. In the first stage, the system based on OptiTrack was constructed, then the site was calibrated and the accuracy test was completed. The accuracy test shows that the system has a sampling rate of 240 FPS and a spatial resolution of sub-millimeter.In the constructed system,open-field tests were carried out on the rats of normal state and the spontaneous activities of rats was tracked. The accurate motion capture data was obtained.According to the obtained data, the model was established and optimized, and the corresponding software for rat behavior pattern recognition was developed. In the second stage, the system completed in the first stage was applied to the behavioral analysis of rat models of fatigue and depression.Rat models of fatigue and depression were made, then tested in the system.Accurate motion capture data was obtained, and behavioral indicators of different states were compared and analyzed. An effective distinction between fatigue and depression, normal and depression was achieved. In this thesis,a high-precision quantitative characterization method of rat behavior is constructed. On the one hand, the large-area open field provides sufficient free space for the spontaneous activity of animals;on the other hand, the three-dimensional motion capture system based on optical markers can be used for recording the free behavior of animals in large fields continuously and completely.Through the high-precision quantitative capture of the behavioral process, accurate measurement of various animal behavioral indicators can be achieved, and more novel indicators can be explored. It is expected to construct a method for objectification, quantification and standardization of animal state. And the ability to automatically identify behavioral patterns provides a more objective analysis method for animal free behavior.
