Objective: Acute myeloid leukemia(AML)is a heterogeneous group of hematologic malignancies,especially acute myeloid leukemia(CN-AML)with normal karyotype.Despite advances in molecular biology,only a few genetic mutations can be used as prognostic biomarkers and therapeutic targets.In this paper,through the application of online database,the expression profile of CN-AML gene was studied,and key genes were screened out through a series of biological information to analyze their effects on prognosis,so as to provide a basis for further judgment of prognosis and search for new possible therapeutic targets.Methods:GEO online database is applied in this article,the chip is obtained by GPL96 GSE34860,depending on the prognosis of CN-AML samples in R of the downloaded data pretreatment,and apply limma package for different prognosis of CN-AML screening differentially expressed genes between different prognosis group volcanic figure and heat maps,further screening by differences,and enrichment analysis,functional annotation and KEGG pathway analysis,gene protein interaction network diagram,get the stable network,select key genes,uneffective survival analysis.Results: 1.87 genes with significantly different expression were screened,including upregulated genes: CDKN2 C,SERPINF1,PROM1,GYPC,DHRS3,and down-regulated genes: MEIS1,PBX3,PPP1R26 CTNNA1,FAM127 A,etc.2.The gene ontology function and pathway enrichment analysis and pathway analysis of differentially expressed genes were conducted through DAVID software,including several pathways such as cell migration and cell adhesion,which are mainly related to extracellular matrix-receptor interaction and adhesion pathway,it is also involved in four signaling pathways,including transcription disorder,complement and coagulation.3.Draw PPI network diagram,and conduct Hub gene identification through cyto Hubba inserted in cytoscape software,and obtain THBS1,VWF,MMP2,APP,SPARC and other key genes.4.Survival analysis was conducted on the key genes in the obtained three subnetworks,and significant survival curves(P < 0.05)were obtained for THBS1 and VWF.The expression levels of THBS1 and VWF had significantly different effects on the survival of patients.Conclusions:In this paper,through the online database of CN-a key gene screening,enrichment of AML gene expression profile analysis and KEGG pathways and survival analysis,draw THBS1,VWF key genes,and found that the prognosis may be meaningful,it will contribute to the understanding of the CN-AML heterogeneity,may become the prognosis and to find new potential therapeutic targets to provide a certain basis.
