The desert steppe is sensitive to climate change.Climate change will exacerbate the warming and drying of the desert steppe and results in vegetation degradation of various degrees.The Stipa Breviflora is dominant species of desert grassland,and is the high quality forage grass that livestock like.This experiment is focused on the cloning of Stipa Breviflora's flowering related genes and analyzing their spatial&temporal expression differences under the artificial warming and nitrogen addition conditions.The main research results are as follows:1.13 genes' fragments were gained from the cloning and analyzing of 42 transcripts which were screened from the transcriptome data.CRY2-1,FT6 and TOC1 belonged respectively to the PhrB family,the PEBP family and the REC family.2.Under natural grazing conditions,the expression levels of PHYB,CRY2-1,COP1-1,ELF3-3,LHY-2 and ZTL-3 were significantly up-regulated during the differentiation stage of floret apex,while the expression levels of PHYA were up-regulated fluctuantly;at the pollination stage,the expression level of CO-7 and FT-6 increased at the stage of the differentiation stage of floret apex.3.In the same flower development period under N,W and WN treatments,there was a big difference in the expression of each gene.In the glume protuberances stage,PHYA was down-regulated in three treatments,while PHYB,CRY1-2,CRY2-1 and FT-6 were up-regulated in N treatment;in the differentiation stage of floret apex,PHYB and CRY2-1 were down-regulated in three treatments,but FT-6 was down-regulated in N and WN expression levels;in the female stamens forming stage,PHYB was up-regulated under N and W treatments,and was down-regulated under WN treatment;CO-7 was up-regulated under three treatments in the female stamens forming stage,while gene FT-6 was down-regulated under N and W treatments,and up-regulated under WN treatment.4.Under N treatment,the expression levels of PHYB,CRY1-2,CO-7 and FT-6 were down-regulated in the differentiation stage of floret apex and the female stamens forming stage;in W treatment,the expression levels of PHYA and CRY2-1 were up-regulated during the female stamens forming stage,while CO-7 was down-regulated in the differentiation stage of floret apex and the female stamens forming stage;FT-6 was up-regulated in the differentiation stage of floret apex,but was down-regulated during the female stamens forming stage under W treatment;the expression level of PHYB,CRY1-2,CO-7 were down-regulated during the differentiation stage of floret apex and the female stamens forming stage under WN treatment;the expression level of FT-6 was down-regulated the differentiation stage of floret apex,but was up-regulated during the female stamens forming stage under WN treatment.
