Population genetic diversity and genetic structure are important research contents of conservation genetics.In this study,mitochondrial DNA and 12 microsatellite loci were used to analyze the genetic diversity and structure of B.b.ussuriensis and B.b.kiautschensis populations.The research objectives are:1)to reveal the structural characteristics of the nucleotide sequence between Cyt b and 12sRNA genes of bubo mitochondrial DNA,and to explore the feasibility of identifying the population origin;2)to reveal the level of genetic diversity and genetic structure of bubo population,so as to put forward corresponding strategies to protect bubo population.The main results are as follows:1.There were repeats of tRNAGlu-CR CR between Cyt b and 12sRNA genes in the mitochondrial DNA of bubo,and the first tRNAGlu was de functionalized,and two control regions(CR1 and CR2)were highly homologous(the similarity was 68%).The structure of the control area of bubo is similar to that of other owls,but there are only two repeats in Domain I:there are four types of repeats in CR1(a,b,c,d,all 22 bp long),in which b is common to all bubo;there is only one type of repeats in CR2(e,69/70 bp long).2.Mitochondrial DNA(Hd=0.995,Pi=0.0053)and microsatellite loci(Ho=0.734,He=0.792)show that the genetic diversity level of bubo population in China is similar to that in Europe,both of which are very high.The phylogenetic analysis based on mitochondrial DNA(BI and ML)showed that the bubo can be divided into two haplotypes(A and B-C,the genetic distance is 0.01),A subgroup only includes B.bubo ussuriensis,its CR2 repeats are all e-1,B-C subgroup includes B.b.ussuriensis and B.b.kiautschensis,and its CR2 repeats are all e-2.But the data analysis based on microsatellites did not show similar results.The two molecular markers showed significant gene exchange between the B.b.ussuriensis and B.b.kiautschensis.The types of repeat units in the mitochondrial control region of bubo are different among its mitochondrial haplotypes,but they are not related to different geographical population,so they can not be used to identify the origin of the population.The genetic diversity of the populations in B.b.ussuriensis and B.b.kiautschensis is high,but we should continue to protect their living environment,reduce the interference of human activities,and crack down on poaching.The two subspecies did not form a significant genetic structure and could be protected as a management unit.
