Objective:The genetic characteristics of mumps virus in Hubei Province in 2019 were analyzed,and the molecular variation was studied to provide baseline data for the monitoring,prevention and control of mumps,as well as vaccine development in China.Method:Used the Real-time reverse transcription polymerase chain reaction(Real-time RT-PCR)to detect the 8 mumps samples which from Hubei Province in 2019.Virus isolation was carried out for the samples with positive MuV nucleic acid.The target gene(SH gene)and the complete genome of MuV were amplified by reverse transcription-polymerase chain reaction(RT-PCR)and then sequenced.Downloaded the representative sequence of the SH gene and the complete genome of MuV from GenBank database.Sequence editing,alignment,phylogenetic and evolutionary analysis were conducted by using the bioinformatics softwareResults:(1)By real-time RT-PCR,8 mumps samples from Hubei Province in 2019 were all positive for MuV nucleic acid;(2)Among the 8 MuV samples,7 out of 8 were successfully obtained the sequence of SH gene(except MuV030).Based on the SH gene,the results of genotype identification showed that one strain belonged to the genotype F dominant epidemic genotype in China,and six strains belonged to the genotype G which epidemic in some areas of China.(3)In the SH gene,the nucleotide difference between MuV097 and the vaccine strain Jeryl Lynn was 15.2%,and there were 14 amino acid site variations.There was no significant trend in the temporal and spatial distribution of genotype F MuV in China.(4)The six strains of genotype G MuV in this study were located in the same evolutionary branch Lineage1 as the genotype G MuV strain in Liaoning Province,Fujian Province and Jiangsu Province from 2011 to 2013,while the genotype G MuV in Shaanxi Province were located in another evolutionary branch Lineage2.In this study,the specific amino acid site of six genotype G MuV samples based on SH gene was T28L.(5)2 out of the 8 MuV samples were successfully isolated and the complete genome sequence was obtained.(6)In this study,MuV012 and MuV097 based on the complete genome were consistent with the topological structure of the phylogenetic tree constructed by each protein.Analysis of the genetic distance of the complete genome and each protein showed that the variation of SH protein is the largest among the seven proteins encoded by MuV,while that of N protein is the smallest,which is relatively conservative(7)Compared with the vaccine strains,MuV012 and MuV097 in this study lacked three N-glycosylation sites,respectively located in P protein 258(NISN),HN protein 127(NCST)and L protein 1727(NLSS);A new glycosylation site 464(NCSG)was added in HN protein of MuV097,and the glycosylation sites 464(NCSG)of HN protein and 1729(NSSE)of L protein were found in MuV012.Conclusions:(1)Following local epidemics in Liaoning Province,Fujian Province,Shaanxi Province,Jiangsu Province and Beijing in 2011~2016,the prevalence of MuV G genotype was detected again in Hubei Province in 2019(2)The analysis of the complete genome of MuV confirmed the high variation of SH protein and its role as a standard molecular basis for genotyping.(3)In this study,the genetic characteristics of MuV SH gene and complete genome were analyzed and the rule of variation and change was described,providing baseline data for the prevention and control of MuV and the development of vaccine in China.Figure[15]table[10]reference[63]...
