DNA nanotechnology is a new technology that based on the physical and chemical characteristics of DNA and combined with modern nanotechnology and molecular biology technology,to build a new controllable and intelligent nanostructure or nanomachine.Structural DNA nanotechnology has received a lot of attention in the field of DNA nanotechnology because of its precise manipulation and diversified assembly methods.And it has been applied in the fields of directed material assembly,biocatalysis,DNA computer,disease diagnosis and drug transportation.In this paper,the research of DNA nanostructure in biological analysis and biological simulation was carried out.DNA tetrahedron is a kind of DNA nanostructure with high rigidity and excellent biocompatibility.The CTDN probe was constructed by combining DNA tetrahedron structure with catalytic hairpin assembly amplification strategy and used for intracellular miR-21 imaging.CTDN probe enters cells by endocytosis without the help of transfection reagent,and has outstanding biocompatibility and enzyme resistance.At the same time,based on the signal amplification strategy of catalytic hairpin assembly,CTDN probe has high sensitivity and selectivity.DNA-AuNPs nanostructures is a type of DNA nanostructures formed by the interaction of DNA and AuNPs through gold sulfur bond or non covalent bond.Five different DNA molecular machines(M1,M2,M3,M4,M5)were successfully constructed through the assembly of DNA and AuNPs.Each machine was powered by entropy driven chain replacement reaction.Based on the communication between DNA molecular machines,three kinds of chemical reaction networks are constructed:cascade regulation network,positive feedback regulation network and negative feedback regulation network.We hope that these two works will provide new ideas for the further development and application of DNA nanostructures.
