The Remorin protein is a plant-specific oligo filamentous family protein that is localized in the membrane domain along with the plasma membrane/lipid and it plays a vital role in plant growth and development,signal transduction and stress response.In this study,'Tsuda'turnip was used as experimental material,the REMORIN genes were cloned,their expression and function were analyzed.The study results are as follows:In this study,we cloned three REMORIN genes from Brassica rapa.The full cDNA sequence of the BrREM1.2 gene and BrREM1.4 containing open reading frames of 585 bp and 618 bp encoding protein of 194 amino acids and 205 amino acids..BrREM1.2 and BrREM1.4 have both Remorin-C terminal domain and Remorin-N terminal domain.The full cDNA sequence of the BrREM16.5 gene containing an open reading frame of 588 bp encoding a protein of 195 amino acids.The BrREM6.5 gene only have the Remorin-C-terminal domain.All there REMORIN differently expressed in the Brassica rapa.The expression of the three genes has tissue-specific.The highest expression levels of the BrREM1.2 could be reached in red root bark,the highest expression levels of the BrREM1.4 and BrREM6.5 could be reached in petals.In the process of anthocyanin synthesis in hypocotyl of turnip seedlings,the expression of three BrREMORIN genes upregulated under high temperature,low temperature,mannitol and ABA treatments,but they were inhibited after PEG,NaCl,KCl,Na2CO3,NaHCO3,IAA and SA treatments,except BrREM6.5 in NaHCO3 treatment.BrREM1.2,BrREM1.4 and BrREM6.5 gene overexpression vectors were constructed through Gateway technology.These vectors were subsequently introduced into Agrobacterium tumefaciens GV3301,so as to obtain transgenic tobacco which overexpressed BrREM1.2 and BrREM1.4 genes.Transgenic tobaccos,which overexpressed BrREM1.2 and BrREM1.4 genes,promoted the growth and development of the plant,inhibited its reproductive development,reduced hundred grain weight,and even caused abortion.BrREMORIN genes can promote anthocyanin synthesis in plants,and BrREM1.2 gene is involved in the response to salt and alkali stress.
