Biodegradable grease technology is widely used in wastewater treatment.In China the concentration of grease in the kitchen waste water is slightly high,and the grease removal rate of the conventional sewage treatment system is not effcient,after collection those water,resulting in excessively high levels of grease in the waste of the kitchen waste.At present,domestic and foreign scholars have not studied much about high-efficiency oildegrading bacteria,and there are few reports on the actual treatment and application of oily wastewater.Therefore,it is very practical to study the degradation effect and application of high-efficiency oil-degrading strains.The water sample from the cafeteria of Anhui Agricultural University in this study.Through screening and purifying strains,ten strains with good lipase production were initially obtained,and then by re-screening and degradation of fat-degrading capacity and lipase activity determination,ultimately a strain C1 with fat-degrading ability was further separated and screened.After morphological observation,physiology and biochemistry Experiments and 16 S r RNA homology analysis determined that the strain was Acinetobacter johnsonii.In this paper,the growth and degradation characteristics of strain C1 were studied.In the experiment,the degradation characteristics of strain C1 were studied by changing the inoculum amount,temperature,p H,shaker speed,salt concentration and soybean oil concentration of the culture medium.Optimal degradation conditions.The results showed that the best conditions for degrading oils and fats were 3% inoculation amount,culture temperature 30?,p H 7.0,shaker speed 180 r/min,optimal sodium chloride salt concentration 2.5%,and fat concentration 4mg / m L.Under the optimal conditions,the rate of degradation of oils and fats reached 85.32%.Tolerance of strain C1 to different concentrations of edible soybean oil was studied in this paper.The strain C1 was able to tolerate a fat concentration of up to 8 mg / m L,and the fat removal rate was still 46.21% at a concentration of 8 mg / m L.Strain C1 was applied to the actual cooking oil waste water.The results showed that in the actual treatment of oil waste water,when the strain treated the oil waste water for 36 hours,the oil degradation rate reached 74.11%,and the COD removal rate in the oil waste water was 71.58%.Continue Over time,the degradation rate of fats and oils increased slowly.At 60 h,the fat degradation rate was 74.52%,the COD removal rate was 72.41%,and the lipase activity value was 9.81 U / m L at 42 h.With the increase of time,the lipase activity value of the metabolic strain of the strain decreased.At the same time as studying the process of oil degradation,the change of amylase over time was also studied.With the change of time,the amylase activity of the metabolism in the wastewater to which the target strain was added also changed in a curve,and the starch at 30 h The enzyme activity value reached a maximum of 6.83 U / m L,after which the enzyme yield gradually decreased with the extension of the treatment time.It can be known from the above studies that the strain C1 is a strain that can tolerate high-concentration oleobacteria,and has a good application prospect in the biological treatment of oil and fat wastewater.
