The ocean occupies about 70%of the earth's surface,and the microbial communities in the ocean sediments play an important role in the biogeochemical cycle.The Woeseiaceael/JTB255 group has the characteristics of wide distribution and high abundance in global marine habitats,and is the core group of marine sediments.This group may play a major role in the global biogeochemical cycle.Currently,the Woeseiaceae/JTB255 group has only one effectively published species,lacking culturable strains.The article takes the Woeseiaceae group as the object for research,and the results are as follows.1.Based on 16S rDNA amplicon sequencing technology to study the distribution and relative abundance of Woeseiaceae in the offshore sea of Weihai and Yuncheng Salt Lake,it was found that the relative abundance of Woeseiaceae was higher in marine sediments than seawater and sample that collected form Yuncheng Salt Lake,and it gradually decreases with increasing sediment depth.In environmental samples,there is a positive correlation between Woeseiaceae and certain microbial groups.A comparative analysis of the genomes of Woeseiaceae.Found that Woeseiaceae lived in a variety of nutritional ways,preferring to grow with oxygen.2.According to the analysis results of Woeseiaceae genomes,design four new medium.Taking the offshore sediments of Weihai and Yuncheng Salt Lake as samples for isolated and cultured Woeseiaceae.A total of 619 strains of bacteria were isolated based on four culture media,distributed in 7 phylums and 149 genera.After the redundant strains were excluded,475 species were isolated,including 275 potential new bacteria(based on<98.6%16S rRNA gene similarity),and the probability of new bacteria was 57.8%.Eighteen species belonging to Chromatiales were isolated through M4 medium.Among them,strains C5,8,24 and XN279 belonged to Woeseiaceae,which expanded the cultivable range of the bacterium.The 16S rDNA similarities between strains C5,8,24,XN279 and Woeseia oceani XK5T were 91.3%,90.7%,91.0%and 90.8%,respectively.3.Genome sequencing of these four strains,and have obtained four genomes.The genome of strain C5 is a 3146397 bp circular structure with a GC content of 67.2%.The genome size of strain 8 is 3115264 bp,contigs are 87,and the GC content is 69.1%.The genome size of strain 24 is 3295170 bp,contigs are 56 and GC content is 66.5%The genome size of strain 279 is 3130633 bp,contigs are 39,and the GC content is 67.8%.From the genome annotations,it can conclude that the growth of Woeseiaceae requires oxygen.It is confirmed through preliminary data analysis that Woeseiaceae plays an important role in the biogeochemical cycle of the marine environment4.Three novel bacteria isolated using the new culture media were identified by polylphasic taxonomy.Strail WTE16T is presented as a representative of a novel species,for which the name Marinilabilia rubra sp.nov.is proposed.Strain 12N15T represents a novel species of the genus Jannaschia,for which the name Jannaschia formosa sp.nov.is proposed.Strain 1E403T represents a novel species of the genus Flavobacterium,for which the name Flavobacterium cerinum sp.nov.is proposed.
