Under stress,polyamine oxidase can oxidize polyamine to generate H2O2.H2O2is an important signaling molecule in plant response to stress.Many studies have shown that apoptosis signals under plant stress are mediated by H2O2produced by polyamine oxidation.In this study,based on the whole genome data of different cotton varieties,the PAO family genes were screened and identified,and bioinformatics analysis was performed to select the gene PAO11 which has the function of participating in stress resistance in upland cotton?Zhongmian 79?and its function was studied.The main experimental results are as follows:Ultra high performance liquid chromatography was used to determine the polyamine content of 79 roots,stems,and leaves of upland cotton in more than 800 sets of stress-treated cotton and exogenous polyamine treatment to alleviate cotton stress Stress,while using D-Arg and DADD to regulate the synthesis and inhibition of endogenous polyamines to study the dynamic changes of the content of polyamines?PAs?during the stress resistance of cotton.Polyamine oxidase can regulate the level of endogenous polyamines under the stress of cotton by oxidizing polyamines.Studying the mechanism of polyamine oxidase is conducive to the use of polyamine oxidase gene expression ability Breeding and breeding.Combined with bioinformatics analysis,and under the conditions of salt stress and simulated drought stress,a polyamine oxidase Gh PAO11 was selected and cloned from the cotton genome.The c DNA of Gh PAO11 is 1799bp in length and encodes 576amino acids.Further,by constructing a eukaryotic expression vector,Agrobacterium tumefaciens GV1301 mediated transformation of wild-type Arabidopsis thaliana to obtain Gh PAO11 transgenic plants.Gh PAO11 was transformed into wild-type Arabidopsis thaliana and T1 generation transgenic plants were obtained.UPLC technology showed that the content of polyamines in transgenic Arabidopsis was higher than that of wild-type,and the expression of Gh PAO11 and some other related genes was higher than that of wild-type under stress conditions.,Indicating that Gh PAO11 is involved in the regulation under stress.Gh PAO11 affects the stress tolerance of cotton by regulating the changes of polyamines under stress,and affecting the content of H2O2,SOD,ABA,etc.in the plant.
