Part 1 lung cancer risk factors in the Chinese population: A Meta-AnalysisPurpose: To explore the risk factors of lung cancer in Chinese population in recent 10 years,and to comprehensively estimate the relative risk of each factor,so as to provide scientific basis for the formulation of prevention and treatment measures for lung cancer.Materials and methods: We searched the database of "CNKI","VIP","Wanfang","Pub Med","Cochrane","Embase" published at home and abroad from 2010 to 2019,the literature of case-control studies on the risk factors of lung cancer in Chinese population were screened,According to the inclusion and exclusion criteria,two evaluators independently carry out data extraction and quality evaluation,and finally use Rev Man5.3 software for Meta analysis.Results: At first,286 articles were searched according to the retrieval strategy,and 17 articles were selected strictly according to the inclusion and exclusion criteria.Finally,17 articles were included in the Meta analysis,with a total of 8521 cases in the lung cancer group and 10154 cases in the control group.The Newcastle-Ottawa(NOS)score of 7 studies was 7,and the NOS score of 10 studies was 6.The relationship between lung cancer and 7 influencing factors including smoking,passive smoking,family history of tumor,home decoration within 10 years,mental and psychological factors,history of respiratory disease,cooking oil fume and lung cancer were studied.Six of them were risk factors for lung cancer.The order of Odd Ratio(OR)from high to low was as follows: family history of tumor(2.86),smoking(2.52),mental and psychological factors(2.34),history of respiratory diseases(2.19),passive smoking(2.12),cooking fume(1.86).Publication bias and sensitivity analysis suggest that the results of this Meta analysis are relatively stable and reliable.Conclusion: Family history of cancer,smoking,mental and psychological factors,history of respiratory diseases,passive smoking and cooking fume are the risk factors of lung cancer in Chinese population.Among them,Among them,the family history of cancer is at the top,and smoking,as a well-known factor inducing lung cancer,remains at a high risk in the Chinese population.Part 2 Identification of nine key genes by bioinformatics analysis for predicting poor prognosis in smoking-induced lung adenocarcinomaPurpose: To screen and identify key genes related to the occurrence and development of smoking-induced lung adenocarcinoma(LUAD),using a variety of bioinformatics analysis tools.Materials and methods: We obtained data for LUAD and normal lung tissues in smoking and non-smoking groups from the GEO chip dataset GSE31210.The differentially expressed genes(DEGs)of LUAD and normal tissues in the two groups were screened by GEO2 R and Venn chart software.The protein interaction network of DEGs was constructed by bioinformatics tools STRING and Cytoscape.Finally,key genes were screened.The overall survival time of patients with the key genes was analyzed by Kaplan–Meier method and expression of screened genes was identified by GEPIA.GO and KEGG bioaccumulation was calculated by DAVID.Results: The lung tissue data of 246 cases provided by GSE31210 data set were analyzed,637 and 336 genes with 4-fold difference in expression were screened in the smoking and non-smoking groups,respectively.The results of the constructed PPI network showed that the modules with the highest scores in the smoking group and the non-smoking group contained 38 and 11 key genes,respectively.Among them,nine key genes were coexpressed in the smoking and non-smoking groups and 29 key genes were specific to the smoking group.Kaplan–Meier analysis indicated that 24 of the 29 genes in the smoking group were associated with worse prognosis.and all of these 24 genes were highly expressed in the smoking group.Twenty-three genes were highly expressed in LUAD and confirmed by GEPIA.GO functional enrichment analysis showed that 9 of the 23 genes were involved in the biological process of smoking-related lung adenocarcinoma,including cell division and mitosis.KEGG pathway analysis showed that BUB1?BUB1B?CDC6?CDC25C was enriched in cell cycle-related pathways,in which BUB1?BUB1B?CDC25C was concentrated in G2/M phase.Conclusion: The integrated bioinformatics method can be used to analyze the relationship between smoking and development of LUAD.Twenty-three key genes were screened out and strongly correlated with adverse prognosis of LUAD induced by smoking.among them,nine key genes(TPX2?MYBL2?CDC6?NCAPH?NCAPG?BIRC5?BUB1?BUB1B?CDC25C)are involved in the biological process of lung adenocarcinoma.
