Circadian rhythm disorder seriously affects human health,and in the process of animal husbandry production,circadian rhythm disorder will affect the normal growth of animals and the occurrence of related diseases.Circadian rhythms are controlled by photoperiods,and studying the effects of different photoperiods on animals could help to uncover human diseases caused by circadian rhythms.Using zebrafish as the research object,this study will be 4 months zebra fish are farmed in normal day-night 20 d environment and all the dark environment,the anatomy of the separation of the liver,the transcriptome sequencing analysis,extract the intestinal contents sequencing microbial flora analysis,study the circadian rhythm disorders affect gut microbes and liver gene expression.The results showed that the swimming distance of the Dark group(all Dark)was significantly lower than that of the control group(P<0.05)after 20d culture.Intestinal microbial flora sequencing showed that the dominant intestinal flora changed in different light production environments,and the abundance of harmful bacteria increased.The abundance,distribution and quantity of each bacterium in the Dark group were different from those in the control group,especially the proportion of Bacteroidetes and Firmicutes in the intestinal tract was significantly reduced.Cryla,Per2,Perla,gpx3,and ggtlb genes were significantly different in liver(P<0.05),and gene function was mainly related to carbohydrate and lipid metabolism and immunity.The results showed that the disturbance of circadian rhythm would cause the changes of intestinal microbial flora of zebrafish,decrease of probiotics,increase of harmful bacteria and decrease of digestive function.It is worth noting that the decrease of liver metabolism affects intestinal flora homeostasis and intestinal flora disturbance further increases the risk of liver disease.The purpose of this experiment is to preliminarily explore the mechanism of circadian rhythm disorder affecting liver metabolites and intestinal microorganisms,so as to provide reasonable photoperiod guidance for production practice,optimize the breeding mode,and improve the utilization rate of natural resources.
