The subfamily Acroneuriinae(Perlidae)belongs to the Systellognatha of Arctoperlaria.For a long time,the phylogenetic relationship within the subfamily Acroneuriinae was not clear,in order to solve this problem and reconstruct the phylogenetic relationships in Acroneuriinae,this study sequenced and analyzed 30 samples from four genera of Plecoptera based on coxl and 18SrDNA,the genetic distance between samples was calculated;we use NJ,ML,BI methods to construct the phylogenetic relationships based on the coxl dataset,18SrDNA dataset and cox1+18SrDNA dataset.The main results of this part are as follows:1.Based on coxl dataset,at the genus level,the monophyly of Sinacroneuria,Flavoperla,Brahmana,and Kiotina are not supported by the NJ tree,the ML tree and the BI tree.Based on 18SrDNA dataset,at the genus level,the monophyly of Brahmana is supported by the NJ tree,ML tree and BI tree.But the monophyly of Sinacroneuria,Flavoperla and Kiotina are not supported by all the trees.Based on coxl and 18SrDNA dataset,at the genus level,the monophyly of Sinacroneuria,Flavoperla,Brahmana,and Kiotina are not supported by the NJ tree,the ML tree and the BI tree.2.The Flavoperla biocellata mitochondrial genome was 15,805 bp long,the whole mitochondrial genome,the PCGs,tRNAs and rRNAs have very similar A+T contents:67.2%,66.4%,70.4%,and 70.6%,the mitochondrial genome contains 37 genes,and eleven PCGs began with the standard start codon ATN(ATT or ATG),however,nad5 used GTG as the start codon,while for nadl it was TTG.Eleven PCGs of the mitochondrial genome had complete termination codons(TAA or TAG),but cox1,cox2 terminated with an incomplete stop codon T.The dihydrouridine(DHU)arm of trnSer(AGN)was reduced to a small loop while the other tRNA genes have the typical cloverleaf secondary structure3.The Leuctra fusca mitochondrial genome was 14,585 bp long,the whole mitochondrial genomes,the PCGs,tRNAs and rRNAs have very similar A+T contents:66.1%,65.0%,68.9%,and 70.6%respectively,the mitochondrial genome contains 37 genes,and eleven PCGs began with the standard start codon ATN(ATT or ATG),however,for nad1,TTG was the start codon,while for atp6 it was GTG.Ten PCGs of the mitochondrial genome had complete termination codons(TAA or TAG),but cox1,cox2 and nad5 terminated with an incomplete stop codon T.The dihydrouridine(DHU)arm of trnSer(AGN)was reduced to a small loop while the other tRNA genes have the typical cloverleaf secondary structure.4.The results indicate that the mitochondrial data can separate the two suborders Arctoperlaria and Antarctoperlaria,supporting their monophyly.The monophyly of Nemouridae,Perloidea and Pteronarcyoidea are also supported.However,the ML trees rebuilt by the PCG13+rRNA dataset of Flavoperla biocellata and the PCG13+rRNA dataset of Leuctra fusca suppoted the paraphyly of Perloidea and Pteronarcyoidea.
