Life cannot be separated from proteins,which are the material basis of life characteristics and have the functions of catalysis,transportation and storage,immune protection and so on.The way in which life bodies exist is that the chemical components of these proteins constantly renew themselves.The process of self-renewal needs protease tools.Protease is an important tool for cells to regulate specific proteins and eliminate misfolded proteins.Protease is an important tool for life to maintain homeostasis,maintain cell structure and adapt to environmental mutations.AAA protease family,a membrane binding protein control system,is a protease dependent on ATP,metal Zn2+.It has both protease activity and molecular chaperone activity,and is able to treat bacteria and chloroplasts,Mitochondria and other membrane proteins were repaired and degraded.The FtsH protease?Filamentation Temperature-Sensitive H?in the large family of AAA protease is an important protein encoded by the ftsH gene.It is involved in the normal metabolic growth regulation,stress reaction,environmental stress and protein balance control and so on in the living organisms.Using Streptococcus mutans UA159 as the main body,we studied the FtsH protease in the AAA protease family,The main results are as follows:?1?in the strain Streptococcus mutans UA159,ftsH knockout mutant could not be obtained normally,indicating that ftsH may be a necessary gene.The growth of ftsH was dependent on the addition of xylose,which confirmed that ftsH was a necessary gene.?2?The Streptococcus mutans UA159 was mutated by ARTP and the mutated strain was knocked out by ftsH.FtsH knockout mutant S517 was successfully screened.The results showed that the?Suppressor?mutation of the genetic suppressor containing ftsH was found in S517.?3?The whole genome of S517 was sequenced and the mutation of suppressor was located in VicR?SMU1517?by SNP analysis and retransformation experiment.?4?vicR gene encoding response regulator?response regulator?,and vicK constitute a two-component signal transduction system,and VicRK is the only necessary two-component regulatory system in Proteus.This study revealed for the first time that there is a close genetic relationship between VicRK bicomponent signal transduction system and ftsH protease regulation system.Based on these findings,we propose the hypothesis that ftsH is involved in the regulation of cell growth,which indicates the direction for the final elucidation of the regulation mechanism of FtsH-like protease.
