Mink pulp cell hyperplasia,caused by Aleutian Mink Disease Virus(AMDV),poses a threat to the development of the animal fur industry.Neutralizing anti-AMDV antibodies can cause persistent infection but is not resistant to AMDV virulent attacks.To date,no specific method has been developed to prevent or cure this disease.In order to eliminate the plasmacytosis of leeches,antibody detection technology screening AMDV-positive leeches has been widely used.This experiment aims to eliminate the cumbersome purification process and expensive cost to prepare recombinant protein antigen,and provide a basis for the detection of AMD and the preparation of AMD colloidal gold test paper.This study was selected for the preparation of protein antigens established by Kai Yu Master's thesis,and the VP2 whole-genome protein of AMDV-G strain was eukaryotically expressed.After purification,polyacrylamide gel electrophoresis showed a molecular weight of 76 kDa;Western blot showed reactivity.The comparison test was carried out by genetic engineering antigen and AMDV antigen CIEP method and PCR method.The coincidence results were 92.6%,94% and 87.5%,respectively.Based on the above,4037 serum samples from two mink farms in Shangzhi City were tested by CIEP for eukaryotic expression antigen and virus antigen respectively.The results showed that the coincidence rate was 93.85%,indicating that the genetic engineering antigen prepared in this experiment can be used as clinical convection immunity.The detection antigen of leech Aleutian disease was detected by electrophoresis.
