Trueperella pyogenes(T.pyogenes),also known as Actinomyces pyogenes or Corynebacterium pyogenes,can induce suppurative inflammation by infecting the mucosa and skin of susceptible animals.It can also be mixed with other pathogens.The iELISA that we established can provide a new method for detection the antibody against T.pyogenes antibody.In this study,Next generation sequencing technology and whole-genome shotgun sequencing technology were used to sequence virulent strain TP-2849 and attenuated strain TP4479.According to the whole-genome sequencing results of porcine T.pyogenes strains,the prediction of genomoc island,VFDB,CARD,COG,KEGG,GO and genome circle of TP-2849(NZ_CP029004.1)and TP4479(NZ_CP029001.1)were completed.All above data we obtained had enriched the data of whole-genome of porcine T.pyogenes strains.The results of prediction of the Virulence Factors Database(VFDB)shows there are five kinds of HSP100/Clp genes in porcine T.pyogenes virulent and attenuated strains.In this study,the BALB/c mice were challenged with the TP-2849 strain and the TP4479 strain respectively.Five kinds of HSP100/Clp genes were detected in seven kinds of tissues and blood of BALB/c mice by the real time quantitative PCR.The result shows that the different expression of five kinds of HSP100/Clp genes in the TP-2849 strain and the TP4479 strain were extremely significant.The result of bacterial growth curve shows the lag of attenuated strain(TP4479)is longer than the virulent strain's(TP-2849),and the stationary of the virulent strain is longer than the attenuated strain's.In conclusion,we can infer that the virulence and the growth rate of porcine T.pyogenes strain are closely related to the transcriptional Levels of five kinds of HSP100/Clp genes.
