In situ screening of saline-alkali-tolerant petroleum hydrocarbon-degrading bacteria can be used to remediate the saline-alkaline soils contaminated with petroleum.In this study,polycyclic aromatic hydrocarbon degrading strainswereisolated from the oil-contaminated soil of the Momoge wetland.The degradation characteristics of the phenanthrene(PHE)by the isolated strain were systematically studied,which provided a good solution for the treatment of wetland petroleum pollution.In this study,the polluted soil in Jilin Oil field was used as samples.The physical and chemical properties of the soil and the microbial species in the fresh soilwere detected and analyzed.Define the growth environment of microorganisms in the region.Trough the analysis of the microbial species,screen the saline-alkali-tolerant PHE degrading bacterium.Using phenanthrene as the sole carbon source,a high-efficiency PHE degrading bacteria resistant to salt and alkali were isolated and identified through morphological and 16 S rRNA gene sequence analysis.This strain was tested in a series of PHE degrading experiments.The residual amount of Phe in culture was determined by gas chromatography-mass spectrometry(GC-MS),and the response characteristics of PHE-degrading strain to salt concentration,pH,initial Phe concentration,bacterial cell density,cultural temperature and incubation time were studied.The suitable growth conditions of PHE degrading strain were obtained.The results showed that three saline-alkali-tolerant PHE degrading bacteria S1-8,S2-1,S3-1 were isolated successfully in this study.The morphological and phylogenetic tree analysis indicated that three isolated strains belonged to Halomonas.All strains had a wide range of adaptation to salt concentration and pH,respectively.Therefore,screening of saline-alkali-tolerant petroleum hydrocarbon-degrading bacteria have a potential application prospect in bioremediation of oil field pollution.
