Oudemansiella raphanipes,the edible and medical fungus,belongs to the Basidiomycota,Agaricomycetes,Agaricales,Physalacriaceae,Oudemansiella.In the present study chemical components of fruiting body of O.raphanipes were isolated,purified and identified,the biological activities of its crude extracts were analyzed as well.Nine of twelve compounds,ergosterol?1?,nicotinamide?2?,uridine?3?,adenosine?4?,diisobutyl phthalate?5?,methyl stearate?6?,D-mannitol?7?,?22E?-23-methyllergosta-5,7,22-trien-3?-ol?8?,butyl phthalate isobutyl ester?9?,were isolated and identified for the first time from the fruiting bodies of O.raphanipes by multiple chromatographic methods.In addition,GC-MS technology was used to analyze the volatile components of the fruiting body of O.raphanipes.A total of 40 compounds were identified.They are mainly aliphatic hydrocarbons,fatty acids,aldehydes,ketones and esters which accounted for 73.063%of the total volatile components.Among them,the highest relative content was Hexanoic acid?13.185%?,followed by Hexadecane?8.048%?,Tetradecane?5.765%?,2,6,10-trimethyl-Tetradecane?5.643%?,and tert-Hexadecanethiol?5.641%?.The evaluation of growth rate was used for determining the antifungal activities of extracts against 13 pathogens,including Rhizoctonia solani,Cylindrocarpon destructans and Fusarium solani.The ethanol extract had strong inhibitory activity against Rhizoctonia solani at concentration 100 mg/mL with the inhibition rate of 52.24%.After a coarse separation derived from ethanol extract,the petroleum ether extract had the highest inhibition rate of 52.90%at 50mg/mL.CCK 8 method was employed to test biological activities of crude extracts including the petroleum ether,and ethyl acetate in four cell lines?listed?in vitro.The results showed that both of petroleum ether and ethyl acetate extract had anti-tumor activities in dose dependent manner.The activities of petroleum ether extract was higher than the ethyl acetate extract.The highest inhibitory effects of petroleum ether extract at 320?g/ml on NCI-H460,HepG-2 and MCF-7 were72.67%,72.83%,53.69%,respectively.The IC50 values of petroleum ether extract on NCI-H460,HepG-2,MCF-7 and SGC-7901 cells were 236.69,229.61,286.84 and 452.97?g/mL respectively.The IC50 values of NCI-H460,HepG-2,MCF-7 and SGC-7901 cells were 909.67,2875.5,803.91and 749.8?g/mL,respectively.
