White tea is a unique tea in China,with slight fermentation and green with light yellow color,has special taste and quality.In this study,a new kind of "Jinhua" white tea was used as a research material to separate and purify the "Jinhua" fungus on surface of the tea leaves,and to study the phosphate-solubilizing capacity of the fungus,the degradation of the herbicide glyphosateand the effect of the fungus on the metabolites in white tea leaves was analyzed,and the results of the genome framework sequence of the fungus were also analyzed.Results are list as follows:In this study,we used isolated and purified the "Jinhua" fungus from white tea by plate spreading.Observed the morphology of the fungus on both PDA and Czapek–Dox medium.The colonies,hyphae,ascospores and other structures,and characteristics of the strains by Light microscopy and electron microscopy were similar with Eurotium Cristatum.Further confirm that strain was a genus of the genus Eurotium Cristatum by 18 S rDNA and ITS1 sequencing.The addition of tea extract under liquid culture conditions can promote the growth of "Jinhua" fungus.The fungus was found to be capable of secreting acid phosphatase by using chromogenic medium.The pH of the medium decreased significantly along with the time of culture increased.The soluble phosphate increased significantly when using Ca-P as a phosphorus source to culture the fungus,indicating that the fungus has ability to dissolve the insoluble inorganic phosphorus for growth.Further research suggest that the fungus could degrade organic phosphorus herbicides glyphosate.Since concentration of glyphosate in the medium showed significant decrease as the fungus growth.After 7 days of culture,the concentration of glyphosate decreased by about 50% and the degradation rate of higher than 70% in 13 days.The experimental results show also that the sfungus could adapt to the higher concentration of glyphosate.By comparing the metabolites of the inoculated and non-inoculated tea leaves with the fungus,it was found that the white tea after inoculation of the "Jinhua" fungus contained much lower catechin content than non-inoculation treatment,especially the the content of pigallocatechin(GC)was significantly decreased.It indicates that the quality of white tea can be changed with fungus inoculation.The whole genome sequencing showed that the total genome of fungus was 26,984,743 bp.At the same time,a gene with relative to the phosphatase secretion was identified,and the protein composition of this gene is analyzed.These results would provide theoretical basis for the application of the "Jinhua" fungus in phosphate solubilizingand the degrade herbicide glyphosate.In summary,the "Jinhua" fungus isolated and purified from "Jinhua" white tea is a kind of Eurotium cristatum,which has phosphatase activity,could dissolve insoluble phosphate,glyphosate and has certain influence on the content of catechin in tea.The results provide a theoretical support for the production of "Jinhua" white tea.
