The problem of soil desertification is serious in China.Microbial ecological research on biological crust in sandy land will be helpful for fixation controlling and desert ecological restoration. In this study, the soil microbial community structure, diversity and soil physical and chemical properties of mossy crust, bare sand surface, soil below crusts and soil below bare sand surface were measured and analyzed,so as to study the influence of mossy crust on soil microbial distribution, aming to provide theoretical basis desert sand fixation. In this paper,high-throughput sequencing technology and Meiji cloud platform were used to analyze the microbial community composition and diversity.Meanwhile, the physical and chemical factors of soil were determined and analyzed comprehensively. The conclusions reached are as follows:1.The soil nutrient indexes of moss crusts are all higher than those of bare sand surface, soil below crusts and soil below bare sand surface;Under the influence of the crusts, the content of available nutrients in the soil below crusts was higher than that in the soil below the bare sand surface. The content of coarse silty sand, silt and clay in the crusts and soil below bare sand surface is respectively higher than that in the bare sand surface and the soil below bare sand surface.2.Microbial diversity of the soil below crusts is the highest. The microbial community composition of the crusts is significantly different from that of the bare sand surface.Under the influence of crust, the composition of microbial community of the soil below it was different from that of the soil below bare sand surface.Available phosphorus, available potassium, available nitrogen, mucilage, silt and coarse sand are important factors affecting microbial community structure.3.Cyanobacteria and Actinobacteria preponderate in the moss crust and are helpful for structural stability of the crust.Compared with the soil below bare sand surface, the relative abundance of Fimicutes increases significantly, while the relative abundance of Proteobacteria and Saccharibacteria decrease significantly.
