Cell electroporation is a biophysical process in which cells form micropores on the bilayer of phospholipids in the cell membrane under the action of an external electric field,which enhances the permeability of the cell membrane.When electroporation occurs in cell membranes,ions,virus particles,proteins and drug can not normally enter cells through macromolecules of cell membranes.The applications of electroporation include:(1)the study of accelerating the death of cancer cells by electric pulse combined with anticancer drugs;(2)the electrotransfection of DNA;(3)the killing of bacteria by non-thermal effect;(4)the transdermal administration.Because cell electroporation depends on the applied pulse field strength,pulse time,cell size,cell type and the conditions of suspension or surrounding tissues,the best conditions for electroporation of different cells and electrochemical therapy of different cell tissues are often determined by experiments.The main contents of this paper include:1.Based on the experiment of Bio-cell electromagnetics,it is necessary to design a high-power adjustable electric pulse generator.This paper introduces the mode of high voltage pulse generation,the choice of pulse driving circuit and Marx generator,and its working principle,output Control of Pulse Number by Scraping Program of Single Chip Microcomputer,which verifies the feasibility of the design of high voltage pulse source.2.Spatial field distribution of electric pulse source irradiation Considering the influence of the transient electric field distribution of suspension tissue fluid on the electroporation effect of cells,the electromagnetic simulation software MAXWELL was used to simulate the electroporation field needed for the experiment.Under parallel electrodes,the spatial electric field distribution of the observed position of cells depends on the distance between electrodes,parallelism,relative position of cells,and the threshold value of the electric field of specific cell membrane perforation.3.Application of Electric Pulse Source in Biological Effects.Firstly,microglia were stained with calcium ion,and irradiated with high voltage pulse source.The effects of different voltage and pulse width on cell electroporation were observed.The effect of cell electroporation was measured by observing the fluidity of calcium ion inside and outside the cell,i.e.fluorescence gray level.The electric field intensity and distribution under different voltage and parameters were obtained by modeling and Simulation of the actual experimental system.The fluorescence intensity curve of microglia cells under different field intensity was obtained by fluo-3AM dyeing and irradiation test.It was found that the peak fluorescence intensity of microglia cells appeared at the same time under different field intensity,and the intracellular free calcium ion concentration increased first and then decreased.
