Twenty rhizospheric soil samples were collected from five types of forest(Phyllostachys heterocycla cv.Pubescens,Cryptomeria fortunei,Camellia japonica,Fargesia spathacea,Fagus lucida)in Fanjingshan National Nature Reserve(FNNR).The Arbuscular Mycorrhizal Fungi(AMF)spores were isolated by wet-sieving and sucrose centrifugation methods and then AMF species were identified based on morphological characters.Molecular identification of AMF by nested-PCR specific amplification sequence alignment of target region of 18 S rDNA gene.To study the diversity of AMF in different forest types,the results are as follows:1.41 species of AMF spores were isolated,22 species were identified.Including 6species of Acaulospora,2 species of Ambispora,7 species of Glomus,1 species of Gigaspora,1 species of Septoglosmus,1 species of Pacispora,1 species of Funneliformis,1 species of Sclerocystis.14 species identified to genus,5 species unknown.Glomus,Acaulospora and Scutellospora is dominant genus.2.The different soil of five forest type have different AMF species community.There are no common species in five forest,and each forest type has its own unique species.The endemic species of Phyllostachys heterocycla cv.Pubescens forest are Gigaspora margarita and Sclerocystis rubiforme,The peculiar species of Cryptomeria fortunei forest are Glomus citricola and Glomus melanosporum,The unique species of Camellia japonica forest are Acaulospora pustulata,Glomus flavisporum,Glomus reticulatum,Glomus pansihalos,Funneliformis badium,The special species of Fargesia spathacea forest is Acaulospora rugosa,The endemic species in Fagus lucida forest are Acaulospora koskei and Acaulospora excavata.3.The diversity of AMF of five forest types in Fanjing Mountain has the following rules:The less of AMF species richness,the greater the fungal spores density,the higher the dominance of dominant species;The more of AMF species richness,the small fungal spores density,the lower the dominance of dominant species.The spore density of AMF in Phyllostachys heterocycla cv.Pubescens forest is 1104 /100 g,10 species;The Cryptomeria fortunei forest is 762 /100 g,15 species;The Camellia japonica forest is 642 spores /100 g,21species;The Fargesia spathacea forest is 566 /100 g,15 species;The Fagus lucida forest is268 /100 g,20 species.4.41 species of AMF spores(FJSAM3-1 to FJSAM3-41)has isolated from soil,Phylogenetic analysis of the 18 S rDNA AML1-AML2 sequence was used to identify the AMF by nested-PCR.Only three sequences were obtained,GenBank: MK592773,MK592774,MK592776.Two of the spores were identified as Glomus,One spore was identified as Paraglomus.The results showed that AMF were abundant in Fanjingshan National Nature Reserve,and there are a large number of AMF species to be identified.Diversity of AMF is closely related to its primary ecosystem.AMF is one of the important ecological factors to maintain the stability of forest ecosystem.Mutual Selection of AMF and Plants makes the diversity of AMF in different forest types significantly different.
