Background: In the animal kingdom,it can be very diverse on regenerative capacity in different species.The research on the tissues and organs of high regenerative potential animal may provide clues for initiating regeneration program in other animals,especially in human beings.Zebrafish is an excellent emerging animal model which can be used to study development and regeneration for its high genetic operability.MLP protein is involved in multiple biological processes and signal pathways,including cell proliferation and cell migration in a variety of diseases such as oncogenesis.However,the relationship between MLP and zebrafish caudal fin regeneration has not been reported.Methods and Results: In this study,we analyzed the expression change and distribution pattern of mlp in zebrafish caudal fin regeneration with q RT-PCR and in situ hybridization.It indicated that mlp expression up-regulated at 4 hours post amputaion(hpa)and then gradually returned to normal level.In situ hybridization results showed mlp mainly expressed in the wound epidermis and distal blastema of regenerative fin.Then,we prepared anti-full-length MLP polyclonal antibody for western blot and fluorescent immunohistochemistry.The results indicated that MLP expression was up-regulated at 12 hpa,the early stage of caudal fin regeneration.It could be phosphorylated as early as 2 hpa and translocated from the plasma membrane to cytoplasm,mainly distributed in epidermal and blastema cells.Finally,we knocked down the expression of mlp through antisense genetic technique morpholino.Afterwards the caudal fin presented a phenotype of regeneration delay.Conclusions: Our work identified that mlp plays a role in the early stage of zebrafish caudal fin regeneration.We speculate that mlp participates in the early events of caudal fin regeneration,such as epidermal cell migration and proliferation,which also provides a basis for further study of the correlation between mlp and tissue regeneration.
