Pseudomonas aeruginosa is a common pathogenic bacteria in clinical and production,which Contains a complex mechanism of resistance.With the abuse of antibiotics,the drug-resistant bacteria have been selectively evolved and developed.The emergence of multi drug resistant strains leads to the increasingly serious drug resistance of Pseudomonas aeruginosa,which seriously threatens the health of human beings and animals.Surveillance of drug resistance and plasmid mediated ?-lactam resistance gene epidemiology from Partial animal sources of Pseudomonas aeruginosa in Sichuan area were conducted,so as to provide guidance for clinical rational use of veterinary drugs.In this study,233 isolates of Pseudomonas aeruginosa isolated from veterinary clinic and production in Sichuan from 2013~2016 were selected as the experimental subjects,The MIC values of 14 antimicrobial agents were determined.The results showed that the tested strains of the ?-lactam antibiotics have strong drug resistance,resistant to ampicillin,cefotaxime and cefoxitin rate were 100%,resistant rate of cotrimoxazole,ceftriaxone,aztreonam,imipenem,cefepime,ceftazidime,ciprofloxacin,gentamicin,levofloxacin,polymyxin B,Amikacin was 97.85%,93.99%,80.69%,93.13%,57.51%,49.36%,5.02%,12.45%,12.45%,9.87% and3.43%.MDP-PA accounted for 79.83% of all strains,most isolates exhibited multidrug resistance to 7~9 antimicrobial agents.Different degree of cross-resistance existed in P.aeruginosa to different ?-lactam antibiotics,ceftazidime on the lowest cross resistance to ?-lactam antibiotics,cross resistance rate in the range of49.36~68.66%;there are also some cross resistance between ?-lactam antibiotics and non-?-lactam antibiotics,In addition to the high cross resistance rate of compound sulfamethoxazole to ?-lactam antibiotics,the cross drug resistance rates of other non drugs to ?-lactam antibiotics ranged was 3.43~20.90%.ESBLs and AmpC ?-lactamase in P.aeruginosa isolates were detected,The results showed that all the isolates were ESBLs and AmpC ?-lactamase-producing positive strains,MBL was not detected.PCR and PCR-sequencing were used to detect22 resistant genes of beta-lactam,The PCR results were as followed: all the isolates harboured ESBLs genes,and 14 isolates harboured plasmid-mediated AmpC genes,MBL gene was not detected.respectively.TEM,OXA-1,OXA-10,CTX-M-15,and DHA-1 genes postive strains were 233,118,28,21 and 14 strains,respectively.The88 strains only carried one drug resistance gene(TEM).145 strains carried more than2 kinds of drug-resistant genes.the strains which carrying more than 2 kinds of drug-resistant genes has stronger resistance than which of single resistant genes.Eric-PCR was used to analyze the homology of Pseudomonas aeruginosa isolates.The results showed that Pseudomonas aeruginosa was divided into 30 genotypes.Among the 233 tested strains,41 transconjugants were successfully obtained,the rate of conjugation was 17.60%,the ESBLs resistance gene was successfully transferred;the transconjugants of AmpC resistance gene positive was not detected.MIC values and PCR results of replicons showed that resistance plasmids carrying genes encoding ESBLs ?-lactamase could spreaded horizontally among bacterium,by this way beta lactam antibiotics drug resistance could be transferred,and causes the spread of multi drug bacteria.
