In order to provide a theoretical basis for the cultivation of Epimedium pubescens,the effective components?icariin and total flavonoids?of E.pubescens were determined by HPLC and UV spectrophotometry,which grow in Tangjiahe national nature reserve in guangyuan city and Jincheng mountain forest park in nanchong city.The the soil microbial diversity of E.pubescens are also analysed by miseq high-throughput sequencing technology.The results were as follow:1.The effective components?icariin and total flavonoids?in different communities.?1?In the Tangjiahe conservation?A,B and C?,the content of icariin in community A was the highest,and the content of icariin in community B was the lowest.The content of flavonoids in community A was the lowest,while the community C was the highest.?2?In Jincheng mountain forest park,the content of icariin was the highway?GL?> the plank road?ZD?> in the woods?SL?> in the bamboo forest?ZL?.The content of flavonoids was ZL > GL > ZD > SL.?3?In different seasons in the highway,the content of icariin was winter? autumn? spring? summer.The content of total flavonoids was winter?summer? autumn ?spring.2.The soil microbial community in different communities.?1?In the Tangjiahe conservation?community A,community B and community C?,the diversity index?Shannon?was community A?com munity B? community C,while the richness degree?chao?was community C?community A?community B.So the community B had the highest diversity index and the lowest richness.The community C had the highest richness and the lowest diversity.The species composition of soil organisms was further analyzed,which indicated the dominant microflora Shared by the three communities as follows: Acidobacteria,Chloroflexi,Actinobacteria,Nitrospirae,Bacteroidetes,Gemmatimonadetes,Planctomycetes,Latescibacteria.The Proteobacteria,Acidobacteria and Chloroflexi were the most dominant microbial community.The content of chlorobacteria in community A was higher than that in community B and C.In community A,the content of nitrospira and bacteroidetes was less than that in community B and C.?2?In Jincheng mountain forest park,the diversity index?Shannon?was community ZD?community GL? community ZL ?community SL,while the richness degree?chao?was community GL? community ZD?com-munity ZL?community SL.The diversity index of community ZD was the highest,while community GL had the highest richness.The diversity index and richness of community SL were the lowest.The species composition of soil organisms was further analyzed,which indicated the dominant microflora Shared by the three communities as follows: Proteobacteria,Acidobacteria,Chloroflexi,Actinobacteria,Nitrospirae,Planctomycetes,Gemmatimonadetes.The Proteobacteria?Acidobacteria and Chloroflexi were the most abundant.The content of chlorobacteria in community GL and ZD was higher than that in community SL and ZL,which Proteobacteria was less than that in community GL and ZD.?3?In different seasons,the diversity index?Shannon?was autumn?winter?summer?spring.The richness degree?chao?was autumn?winter?summer?spring.The results indicated the changes of diversity index and richness index in the four seasons were consistent.The species composition of soil organisms was further analyzed,which indicated the dominant microflora Shared by the four seasons as follows: Proteobacteria,Acidobacteria,Chloroflexi,Actinobacteria,Nitrospirae,Proteobacteria.The amount of Proteobacteria in spring was significantly lower than that in the other three seasons.The Acidobacteria are most abundant in autumn.The Chloroflexi in winter is less than other three seasons.The Actinobacteria varied little over the four seasons.The Nitrospirae in spring and summer were much more than the autumn and winter.3.The correlation analysis of the rhizosphere soil microorganism and effective components of E.pubescens.?1?The Acidibacter in the soil of Tangjiahe conservation area are significant positively correlated with the content of icariin,while the Nocardioioides,Haliangium and Nitrospira were significant negative correlation with the content of icariin.There was no significant correlation between total flavonoids content and soil microorganisms.?2?The BurkholderiaspyxAZ32 in the soil of Jincheng mountain forest park was significant negative correlation with the content of icariin.There was no significant correlation between total flavonoids content and soil microorganisms.?3?In different seasons,the H16 was significant negative correlation with the content of icariin.the Hliangium was significant negative correlation with total flavonoids content.
