Oryzias is characterized by its small size,strong fecundity,short reproductive cycle,large amount of eggs,continuous laying,high transparency of eggs,sensitivity to water quality,etc,which has been widely used in the research fields of developmental biology,molecular biology and toxicology.The Oryzias curvinotus is widely distributed in the coastal areas of Guangdong,China.Through sampling and investigation of the Oryzias curvinotus,our research group found that its habitat waters fluctuate greatly with the fluctuation of tide salinity,and its adaptability to salinity fluctuation was strong,which has the potential to be used in seawater environmental monitoring.However,the basic research on the gill was insufficient in China,the following studies were carried out in this paper: 1.Continuous stereoscopic observation of the embryonic development of the Oryzias curvinotus;2.The histological changes of different developmental stages of Oryzias curvinotus were observed by serial section technique,and the data of the development of female and male glands were obtained;3.Biomarking of protogerm cells by microinjection technology.And tracer observation.The main results were as follows:1.The embryos of Oryzias curvinotus lasted for 10-12 days at 26?,followed the development model of teleost fish,and went through eight stages..According to the changes of morphological characteristics in each stage,such as the formation of the blastocyst,the number of somatic segments,the occurrence of organs,the size and movement of the embryo body,etc.One step was divided into 35 stages.The spontaneous fluorescence mainly began at the 12-sarcomere stage,and was produced by the substances in the iridescent cell mass under the excitation light.It mainly distributed in the surface of the skin on the back of the brain and spinal cord.2.The gonad of the Oryzias curvinotus lasts for 2-3 months from hatch to sexual maturation.From 5-10 days after membranous hatch,the gonad began to differentiate.The structure of ovarian cavity was observed in the ovary of female fish 40 days after hatch.After 50 days of hatch,mature sperm was seen in the testis.The gonad development of female individual was earlier than that of male individual.The spawning type of Oryzias curvinotus belongs to asynchronous continuous spawning.3.In this study,the sequence alignment analysis of vasa gene between Oryzias curvinotus and Oryzias latipes showed that the consistency of 3'-UTR region reached 86% with high homology.The recombinant plasmid of vasa gene was successfully constructed to label the progenitor cells of Oryzias curvinotus.In this study,the early embryonic development,gonad development,egg and spermatogenesis of the Oryzias curvinotus were recorded,which provided important basic data for Developmental biology and Genetic biology.
