Lectins are non-immunogenic sugar binding proteins that are widely distributed in animals,plants and microorganisms.The most basic property of lectin is the agglutination of red blood cells.Based on this,this study will focus on the changes of lectin and blood action,and then apply image processing techniques to the monitoring of the experimental process.By designing a blood coagulation experiment,a video of the change of agglutination during the experiment is taken and the collected images are processed.In addition,an area ratio analysis method and a gray-scale analysis method are proposed to monitor the reaction process of the experiment.The main contributions of this thesis are shown as follows:1.The collected color experimental images are taken every 100 seconds and converted into gray images to be numbered.Mean filtering and median filtering are used to denoise the image.Compared with the results of filtering,it is found that median filter can better remove noise.Therefore,median filter is used in preprocessing.2.Image segmentation of denoised images.Selecting images in different agglutination states,using edge detection,threshold segmentation,and other methods for segmentation.By comparing and analyzing the experimental results,it is found that the maximum inter class variance method has obtained the best segmentation effect,and it can basically show the agglutination state in the process of the experiment.Binary images are easy to calculate and convenient to store,so the largest variance between classes is chosen as the segmentation algorithm in this study.3.According to the principle and experimental phenomena of the reactivity of the lectin to the blood,the state of the blood cell sedimentation in the agglutination process is regarded as the characteristic quantity of the change.Based on the obtained binary image,an area ratio analysis method and a gray-scale analysis method are proposed.Through the data analysis obtained from multiple experiments,the above two analysis methods can describe the change of agglutination state during the experiment,and the obtained data are stable and accurate.It is proved that this method can be used to dynamically monitor the action process of the agglutinin and blood in the experiment.In this paper,we focus on the observational difficulties in the coagulation process of lectin,based on the agglutination of lectin and blood,combined with image processing techniques,to monitor the macroscopic dynamic changes of blood cell deposition during the experiment.On the basis of the analysis method,the dynamic change curve is depicted.Compared with the naked eye,the image and data are strict.
