Since the realization that cells are the basic unit of life,researchers have been trying to characterize and classify cell types based on cell characteristics,while traditional RNA-seq and gene chip analysis at the population level ignores heterogeneity between tissue cells.So it is difficult to reflect the law of life activitily at the single cell level.Therefore,in order to analyze the heterogeneity,different expression between cells,and even fund new cell types we should analyze from a single cell level.Based on this,technology of single cell sequencing has begun to appear.This paper introduces Microwell-seq—a new single-cell sequencing platform independently developed by our laboratory.Compared with other single-cell sequencing platforms,Microwell-seq has the characteristics of low cost,high throughput and easy operation,which greatly reduces the developement of single-cell sequencing.In this paper,the operation flow of Microwell-seq and the analysis of data are introduced.At the same time,the pancreas and liver of mice are tested and analyzed,which not only classifies the pancreas and liver cells of mice,but also proves the applicability of the Microwell-seq platform.
