Acyl-CoA binding proteins(ACBP),containing acyl-CoA binding domain(ACB),are involved in lipid synthesis and decomposition,acyl-CoA homeostasis and other biochemical processes.Germin-like proteins(GLP)are a kind of seed germination marker with cupin domain.GLPs participate in a variety of physiological and biochemical processes as enzymes,structural proteins and receptors.Both of ACBPs and GLPs play an important role in plant growth and development as well as biotic and abiotic stress response.However,the molecular mechanism of salt-alkali response mediated by these two kinds of proteins are still unclear..Puccinellia tenuiflora is a monocotyledonous plant with strong salt and alkali resistance.It is an ideal material for investigating the mechanism of plant salt and alkali response.In this study,bioinformatics and molecular genetics techniques were used to clone the PutACBP6 and PutGLP2 from.tenuiflora.The length of PutACBP6 open reading frame is 1611 bp,which encodes 536 amino acids.It belongs to the ACBP Class ? subfamily,containing a signal peptide and an ACB domain.RT-qPCR analysis showed that PutACBP6 was expressed in the roots,stems,leaves,sheaths,flowers,spike and seeds of P.tenuiflora.Among them,the expression level in roots,flowers and spike of.tenuiflora was higher than in other tissues,while the relative mRNA level in seeds was the lowest.Under abiotic stresses(200 mmol/L NaCl,100mmol/L NaHCO3,75 mmol/L Na2CO3,10 mmol/L H2O2and 4?),PutACBP6 in roots and leaves showed up-regulated expression pattern.It is suggested that PutACBP6 may be involved in the response of salt,alkali,oxidation and low temperature stress.Transient expression localization analysis in Nicotiana tabacum leaf cells showed that PutACBP6 was located in cytoplasm.The length of PutGLP2 open reading frame is 666 bp,which encodes 221 amino acids.It belongs to the GLP subfamily 2,containing a signal peptide and a cupin domain.Transient expression localization analysis in N.tabacum leaves showed that PutGLP2 was localized to the extracellular matrix.Resistance analysis of PutGLP2-overexpression yeast strain showed that the overexpression of PutGLP2 can increase the resistance to salinity,heavy metals and oxidative stress in yeast strain.This result indicated that PutGLP2 played an important role in regulating the response to abiotic stresses of P.tenuif P.tenuiflora.In summary,PutACBP6 and PutGLP2 may be involved in regulating the response to abiotic stresses of P.tenuiflora.These results enriched the information of plant stress tolerance genes.At the same time,it provides a new evidence for the in-depth study of the regulation mechanism of the stress response in.tenuiflora.
