The cassava(Manihot esculenta Crantz)is a perennial erect shrub or tree.It usually has a plant height of 1.5 m to 3 m.It is also known as arbor,cassava,southern sweet potato,and wood sweet potato.Cassava is a genus of the genus Cassava,which is a high-yielding plant of carbon 3 and carbon 4 plants different from most plants.The edible part of cassava is the root of cassava,which is used as a source of carbohydrates for human consumption and consumption because of its low protein content and high starch content.Due to its high yield and low planting cost,cassava is one of the natural raw materials for animal feed.Because of its high protein content in its leaves and stems,cassava can be used directly as an alternative to the grain part of animal feed.Because of its high starch content,cassava roots are also industrially useful for making industrial products such as alcohol using tapioca starch.Auxin is involved in many plant physiological and biochemical processes,such as cell elongation and division,vascular tissue differentiation,leaf and flower litter,and regulates the plant's apical dominance and tropism.Indole acetic acid has different effects on plants at different concentrations.In the case of low concentrations of indole acetic acid,indole acetic acid promotes plant growth.Conversely,in the case of high concentrations of indoleacetic acid,indole acetic acid inhibits growth and can even lead to plant death.No previous report on the IAA/ARF gene family in cassava has been reported.Is the structure of IAA/ARF in cassava similar to that in other species?Is there a correlation between the IAA/ARF gene family in cassava and plant growth and stress resistance?Based on the complete sequencing of cassava genome,the IAA/ARF gene family of cassava was used as the research object,and all the genes of this family were located in the whole genome of cassava,and related bioinformatics analysis and cloning were carried out.Expression specificity analysis and expression differential analysis under stress to solve the above problems.The main results of this study are as follows:1.A total of 38 IAA gene families and 27 ARF gene families were identified from the cassava genome database,all of which contained intact or partial AUX/IAA conserved protein domains.2.The average molecular weight of the cassava IAA family gene was 26.07 kDa,the average isoelectric point was 6.97,the average molecular weight of the cassava ARF family gene was 86.74 kDa,and the average isoelectric point was 6.29.It can be seen that the protein size of the ARF family is significantly larger than the size of the IAA family.3.The cassava IAA gene appears on chromosomes other than chromosomes 2,10 and 17,and the genes are clustered,and most of the genes are concentrated on certain segments of certain chromosomes.4.Through the phylogenetic tree analysis,two pairs of relatively close genes were identified,naMely Manes.14G153900 and At5g57420,Manes.14G107100 and At2g01200.5.Through the protein interaction network of cassava IAA/ARF,we found that among these matched genes,the corresponding MP and AXR3 nodes of Arabidopsis thaliana are the most important nodes,and the corresponding cassava gene is Manes.10G124800,Manes.11G051200.6.Overall,the IAA/ARF gene family of cassava has several IAA/ARF gene family gene expressions in the main lateral roots of all varieties.Compared with the internal gene family,the expression of individual genes showed great differences.SoMe genes were expressed in the main lateral roots,while some genes were not detected in the main lateral roots,and the same gene was in six cassava.There was also a certain difference in expression between the varieties.7.Under stress conditions,the expression of individual genes showed great differences.Some genes were expressed under various stress conditions,while some genes were significantly inhibited under stress.Some genes were in common condition.There are only a few expressions under it,and the amount of expression will gradually increase with time,which may be the embodiment of negative feedback.8.A T vector containing 11 cassava IAA/ARF genes was constructed and stored in E.coli DH5a.
