The Q gene controls the formation of the seed base in wheat,which is closely related to the seed shattering.In this study,total 252 genotypes with different shattering were identified,and the high-generation lines L40,ASX-14,ASX-4,ASX-9 and ASX-10 with weak-shattering rates were identified,which are:2.67%,2.43%,2.37%,1.93%,and 1.80%,which can be selected as parents with weak-shattering and used in oats breeding for higher yield.In this study,method of homologous cloning was used to obtain the different copies of Q gene in Avena nuda L.for the first time.Total of 6 copies of AnQ were detected in Bayou 12,and which were named as AnQ-Al and AnQ-A2,located in A chromosome;AnQ-C1 and AnQ-C2 located in C chromosome;AnQ-Dl and AnQ-D2 located in D chromosome.At the same time,compared with sequences in different copies of AnQ genes,92 SNPs were detected,and according to specific SNPs located in its chromosome,two gene markers AnQ-A and AnQ-D were developed,which could be amplified specifically AnQ located in A and D chromosomes in Avena nuda L.In order to clear the expression of AnQ gene in Avena nuda L.with different shattering,semi-quatitive RT-PCR was used to analysis.The results revealed that different transcript levels and pattern were detected in genotypes,AnQ-A and AnQ-D had higher transcript level in genotypes with shattering than genotypes with non-shattering.In addition,mis-splicing of AnQ-D was detected in genotype of ASX-9 with non-shattering.
