In eukaryotic cells,vesicle trafficking is the main route for transportation of proteins and lipids.It includes vesicle budding,movement,tethering,docking and fusion.SM(Sec1/Muncl8-like)protein families play an important role on membrane fusion which is mediated by SNARE complexes.The SM protein families are divided into four subfamilies,Sec1 family,Slyl family,Vps33 family and Vps45 family.Each subfamily plays a critical role in a specific compartment.Among them,the Sly1 family functions between the endoplasmic reticulum and Golgi apparatus.Our previous studies discovered SM protein AtSly1 by pull down analysis oftransgenic plants of Arabidopsis thaliana p35S::myc-AtSYP81,but the function of AtSly1 gene is not clear.This studyfurther proved the interaction between AtSly1 and AtSYP81 in Arabidopsis thaliana by Pull-down experiment of pSYP81::GFP-SYP81 transgenic plants.Through the fluorescence localization,we found that AtSly1 is localized to endoplasmic reticulum,Golgi apparatus and cytoplasm.Morphological observation showed that overexpression of AtSly1 affectsseed development,and through the Western-blot testwe found thatthe changes of AtSly1 expression level affect the normal transport of seed storage proteins.
