Tuber indicum is a species with dominant distribution and yield in China.It has high economic value and its morphology is very similar to that of French black fungus.In recent years,it has attracted great attention from domestic scholars.Based on the understanding of the ecological habits of T indicum and mycorrhizal synthesis at home and abroad,the mycorrhizal synthesis of this species with Corylus heterophylla and Picea crassifolia was studied in this article.Morphological and molecular biological methods were used to identify the synthesized ectomycorrhizae.The influencing factors of mycorrhizae morphology were discussed.Through detailed observation and comparison of the morphological and anatomical characteristics of artificial mycorrhiza,It was found that the macromorphology of mycorrhizae formed depends definitely on the hosts,Few anatomical differences were observed between the mycorrhizae formed on the three plants.In addition,Substrates and age of mycorrhizae had influence in shaping the macromorphology of mycorrhizae.Mycorrhiza formed by T.Indian on host plants is highly similar to that of.melanosporum.Through molecular identification of tested mycorrhizae and construction of phylogenetic tree,it is confirmed that the mycorrhizae synthesized in this experiment is Indian truffles,T.Indian are closely related to THimalayan and.melanosporum.For C.heterophylla and P.crassifolia factorial experiment designs were used to screen out the optimum substrate,pH and the amount of inoculum.Statistic analysis indicated that for C.heterophyllathe optimal conditions for synthzing mycorrhizae of T.indicum were:substrates of peat:vermiculite:perlite(4:3:3),pH=7.5,9×106 spores/seedling and for C.heterophylla;P.crassifolia the optimal conditions for synthzing mycorrhizae of T.indicum were:pH=7.5,3×106spores/seedling.This study provides theoretical and applied basis for the protection,development andartificial cultivation of T indicums.
