Wohlfahrtia magnificci is the pathogen of Alxa Bactrin Camel vaginal maggot disease,harming the vaginal part of Alxa Bactrin Camel,and has become one of the main diseases hindering the healthy development of Alxa bactrian camel.In this experiment,the morphological and structural characteristics of Wohlfahrtia magnifica pupa were studied by electron microscope and light microscope.On this basis,the life history of Wohlfahrtia magnifica was studied.The Wohl.fahrtia magnifica at different developmental stages was taken as the research object,high throughput sequencing(Illumina Hiseq4000)was used for transcriptional group sequencing and bioinformatics analysis,and the target gene was verified by fluorescence quantitative technique.The results showed that the development time of larvae and pupa of Wohlfahrtia magnifica was 1-12 or 18 days,and the growth and development of pupa were divided into three stages.The 25 sequencing results shows that the valid data volume of each sample is above 4.90G,the content of the G C is more than 35.35%,the content of the Q20 is more than 97%,a total of 73303 unigenes were obtained.Comparison with NCBI_nr,GO,KEGG,Pfam,Swiss-Prot and eggNOG databases,among them,eggNOG database annotated the most unigenes,35066,distributed in 23 protein functions,among which the translation modification,protein turnover of the unigenes account for a large proportion;Unigenes involved in biological processes and redox processes account for a large proportion of GO functional enrichment analysis(including molecular functions,cellular components,and biological processes,50 branches).Unigenes involved in the signal transduction process account for a large proportion of KEGG enrichment analysis.The above data indicated that the sequencing result of the transcript set was good,which laid a foundation for further research.Through gene differential expression analysis,110 olfactory-related genes and 119 wing development-related genes were screened out.Among them.Further gene annotation found that the expression levels of olfactory-related gene Obp99b?Obp56a?Obp99a?Obp19d and wing development-related gene Bai?Eca?Dpp? were significantly different between groups(|log2FC|>1,P>0.05).The results of fluorescence quantitative per test showed that the difference of Obp99b?Obp56a?Obp99a?Bai gene between groups was significant(P<0.01),the difference between Eca,DppIII genomes was significant(P<0.05),and the difference of Obp19d gene was not significant(P>0.05).In conclusion,Obp99b?Obp56a?Obp99a gene is related to olfactory development and Bai?Eca?Dpp? gene to wing development.Conclusion:(1)The morphological structure of the pupa of Wohlfahrtia magnifica at different developmental stages was supplemented,and the life history of different developmental stages of Wohlfahrtia magnifica was studied deeply.(2)The database of transcription group of Wohlfahrtia magnifiea was established.(3)Obp99b,Obp56a,Obp99a gene was related to olfactory development,Bai,Eca,DppIII gene is associated with wing development.
