To fully understand bioinformations of Brucella Rev.1 vaccine strain,present research conducted genome-wide sequencing for Brucella Rev.1 through PacBio platform.Results shows that a full-length genome with 3,299,187 bp,and GC content ratio reaches to 57.23%,corresponding information of chromosome 1 is 2,121,370 bp and 57.15%,and corresponding information of chromosome 2 is 1,177,817 bp and 57.34%.3318 encoding genes with a length of 2,857,932 bp were annotated,and the average length of each gene is 861 bp with 58.34%GC content.37 scattering and repetitive sequences,89 tandem and repetitive sequences,71 satellite-DNA,54 tRNA,9 rRNA,2 sRNA and 10 gene island sequences were predicted.The amount of annotated genes referred to database of GO,KEGG,COG,NR,TCDB,PHI,VFDB,ARDB and CARD is respectively 2148,3253,2703,3274,390,223,228,1 and 21,which includes 158 secreted proteins and 78 T3SS effector proteins.Gene sequences of BLS,gmd,L7/L12,OMP25,OMP28,P39,VirB10,wboA and Ery of Brucella Rev.1 vaccine strain were aligned with 8 reference strains in Genbank,all sequences showed well homology more than 99.4%.VirB10 of Brucella Rev.l exhibited 1 extra insert of 18 bp compared with reference strains of 16M,M28 and M5-90,and exhibited 2 extra insert of 9 bp and 1 gene deletion of 24 bp compared with reference strains of 2308,RB51 S19 and A19,and exhibited 1 extra insert of 9 bp and 1 gene deletion of 24 bp compared with reference strain of S2.Ery of Brucella Rev.l vaccine strain exhibited 1 extra insert of 705 bp compared with reference strain of S19 and 1 gene deletion of 16 bp compared with reference strain of S2.BLS,gmd,L7/L12,OMP25,OMP28,P39 and wboA showed no regular difference with that of reference strains.
