As an emerging spectroscopy technology,terahertz spectroscopy technology is complementary to infrared and Raman spectroscopy.It has certain application value in the analysis of main component content and component identification of antibiotic drugs,and provides a rapid,safe and effective detection method for the quality evaluation of antibiotics.In this paper,the terahertz characteristic spectrum of 27 antibiotics such as minocycline was firstly detected by terahertz time-domain spectroscopy system,which provided data support for qualitative identification and quantitative analysis of antibiotics.Secondly,the characteristic spectra of four antibiotics were detected by double ternary and two-dimensional liquid chromatograph,and the qualification of antibiotics from different manufacturers was identified,which provided a comparison for qualitative and quantitative analysis.Then,qualitative analysis and quantitative analysis of four types of antibiotics were carried out by cluster analysis,principal component analysis and multiple linear regression.Then,qualitative analysis and quantitative analysis of the four antibiotics were carried out by cluster analysis,principal component analysis and multiple linear regression.Finally,13 kinds of antibiotics were accelerated aging through high and low temperature damp heat test chamber to explore the aging rule of antibiotics.The research indicates:(1)Combining terahertz spectroscopy technology with cluster analysis to have high resolution in identifying the eligibility of antibiotics in different manufacturers.Some antibiotics in the same kind of antibiotics can be effectively classified by clustering methods,and the drug substance and preparation can be distinguished.(2)The terahertz spectroscopy technique has obvious advantages in the quantitative analysis of antibiotics.The correlation coefficients of minocycline hydrochloride capsule,amoxicillin and ofloxacin is above 0.99.The average relative errors of multiple linear regression models are 5.2%,8.9%,10.0%,7.8% and 3.5%,espectively.The correlation coefficient between clarithromycin,azithromycin and metronidazole quantitative models is above 0.98,and the average relative errors of the multiple linear regression model are 11.2%,12.1% and 17.0%,respectively.Due to the strong interaction between the molecules after clarithromycin milling,resulting in poor uniformity of sample preparation,and zithromycin has a consistent sample consistency with increasing sample concentration,and the terahertz characteristic absorption peak of metronidazole is in the high frequency band where power is sharply attenuated.(3)The terahertz spectroscopy technique can be applied to the aging analysis of antibiotics.The absorption coefficients of the terahertz peaks at before and after aging of 13 antibiotics have changed to some extent.Some aging samples can be distinguished by Euclidean distance,and the Euclidean distance is positively correlated with the degree of aging.The longer the aging time,the more antibiotic ingredients,the signal-to-noise ratio of the terahertz characteristic spectrum decreases,and the effective spectrum range decreases.(4)The qualitative and quantitative analysis results of antibiotics based on terahertz spectroscopy technique are consistent with the results of liquid chromatography.The liquid chromatography analysis method can be used to calibrate the terahertz spectroscopy method.This paper proposes the use of terahertz spectroscopy technique to evaluate the quality of antibiotics,and provides a new detection method to ensure the functional effectiveness and safety of antibiotics.
