Alternaria brassicicola can cause black spot in cruciferous vegetables,resulting in reduced yield and quality.This disease also reduced edible safety and huge economic losses.Transcription factors have important regulatory effects on the growth and pathogenicity of fungi.Among them,C6-type zinc finger transcription factors?TFs?are reported in the regulation of sporulation and pathogenicity of various fungi.But little is known of these TFs in A.brassicicola.Study on functons of these TFs in A.brassicicola can widen our knowledge of pathogenic mechanisms,which is helpful to the future treatment of black spot.In this study,A.brassicicola was used as the research object,and the transcription factor gene AB04837 was cloned.Split-marker PCR was used to generate the AB04837 disruption construct.And PEG-mediated protoplast transformation to disruption the gene AB04837 gene deletion mutant?AB04837 and its complement C1 were successfully obtained.The main results are as follows:?1?AB04837 gene has a full length of 2104 bp and a full-length cDNA of 2055 bp,encoding 684 amino acids.It contains a conserved Zn2+domain and a 49 bp intron,which is closely related to P.tritici-repentis.?2?AB04837 gene is involved in the regulation of vegetative growth of A.brassicicola.The color of the mutant strain is white,the aerial hyphae are lusher,and the growth rate is significantly faster than that of wild type.?3?AB04837 gene is involved in the regulation of the production of conidia of A.brassicicola,and the mutant does not produce conidia.?4?AB04837 gene is involved in the regulation of the anti-oxidative stress of A.brassicicola.The growth rate of the mutant in the oxidative stress medium is significantly reduced,and the sensitivity to oxidative stress is enhanced.?5?AB04837 gene is involved in the regulation of the stress response of A.brassicicola,and the gene deletion mutant is more tolerant to low-concentration osmotic stress,and the tolerance to high-concentration osmotic stress is significantly reduced.?6?AB04837 gene is involved in the regulation of the pathogenicity of A.brassicicola,and the pathogenicity is significantly reduced.Disruption of AB04837 gene may reduced the colonization ability of A.brassicicola,but may influenced little on the host penetrating ability.?7?AB04837 gene is involved in the regulation of the cell wall integrity of A.brassicicola,and the sensitivity of gene deletion mutants to SDS,CFW and CR is enhanced.In summary,AB04837 gene is involved in the regulation of vegetative growth,conidia production,stress response,pathogenicity and cell wall growth and development of A.brassicicola.
