Gut microbiota are closely related to human metabolism and various diseases.Numerous studies have shown that Prevotella is not only one of the three enterotypes,but also the core bacteria genus of human gut microbiota.However,the role of P.copri in the human intestinal microbiome is controversial.Some studies suggest that it is related to rheumatoid arthritis,type 2 diabetes,hypertension,autism,etc as a potential pathogen.The others believe that it performs functions beneficial to the health of the host in the human intestine as an indicator of the microbiome related to health status,such as improving glucose metabolism and assisting anti-inflammatory effects.The serious lack of the phylogeny,genetic information and physiological characteristics of the strain hinders the deep exploration and understanding of its relationship with the host.This study attempted to isolate P.copri strains from Chinese intestinal sources and combine genome sequencing and transcriptome sequencing to investigate their specific genetic and physiological characteristics.The results were as follows:(1)By combining 16s amplicon sequencing and anaerobic separation and culture techniques,multiple P,copri strains were successfully isolated,including 2 strains belonging to Chinese endemic subspecies;(2)Besides the genome draft of all isolated strains,the whole genome sequence of P.copri was firstly obtained represented by YF2.We identified the characteristics of the strain through the functional annotation,and specially focused on the copy numbers and heterogeneity of the rRNA operon.(3)For the analysis of carbohydrate-related hydrolase,it is found that P.copri from Chinese specifically contains the alginate-like enzyme(PL6/PL17)encoding gene.Through the transcription analysis and heterologous expression,it was confirmed that YF2 strain can use alginate as carbon source for growth owing to its alginate lyses activity.(4)the differential transcriptional expression profile of YF2 under different pressure conditions(37? anaerobic,25? anaerobic,37? aerobic)are characted by RNA-seq and and analysied by DESeq.On the one hand,this study provides a strong basis for understanding the metabolic characteristics and physiological adaptation of important enterobacteria P.copri from the genomic and transcriptional levels,and on the other hand,it will provide a premise basis for further development of the genetic manipulation method of the bacteria.
