Marek's disease(MD)is a tumor disease caused by chicken marek's virus(MDV),which brings huge economic losses to raise industry every year over the world.People have been committed to the prevention of MD for years,although has obtained some achievements,the pivotal problems havn't solved out.Since"RBC immune" notion was brought up in 1981,there has been more and more researchs working on red cell immune.We also find out that red blood cells not only act as the carrier of gas transportion but also bear the function of many aspects,such as participating in the immune and metabolic processes.At present,researches on red cell immune mainly on human red blood cells,and in terms of nucleated red cells,researches mainly on some fish and reptiles,studies on chicken red blood cells are limited.In this study,we adopted the method of the transcriptome sequencing,selecting differentially expressed genes between the control group and MDV infection group at different development stages(14 dpi and 22 dpi),developping bioinformatics analysis to these differentially expressed genes.For further understanding the infection mechanism of MD from the perspective of red cell immune,analyzed the gens involved in signal transduction and physiological metabolism,discussed their potential relevance to MDV infection mechanism,which will provid a new train of thought for MD control.At the same time,we also analysized the scheduling expression differentiation of the two control group and two MDV infection group to study the role of red blood cells in innate immune overall.Gene sequence results showed that,compared to the control group at 14 dpi(14 days post infection),MDV infection group geted 235 differentially expressed genes,in which 117 up-regulated and 118 down-regulated;compared the control group at 22dpi(22 days post infection),MDV infection group got 218 differentially expressed genes,155 of them were up-regulated and 63 were down-regulated;In the different stages of 14dpi and 22dpi,the control group got 854 differentially expressed genes,among which 319 up-regulated,535 down-regulated;MDV treated group got 53 differentially expressed genes,21 up-regulated,32 down-regulated.We also made an annotion for differentially expressed genes by Nr,Swiss Prot,GO,COG and KEGG database,the vast majority got the comments,in these genes,TLR3?STAT1?LY96?PIK3CD?Fos ? MMP9 involved in TLRs sigal pathway.At last,differentially expressed genes analysis results showed that,the genes relating to immune on red blood cells generally up-regulated in MDV infection group compared to the control group,which indicated that the red blood cells play a certain role in resistting MDV infection and they may be at the forefront of resisting the pathogenic microorganism infection,which provided a new train of thought for the research of MD.
