Brachypodium distachyon belongs to Poaceae family,which has a close phylogenic relationship with the major cereal crops,including wheat,barley and rye.It is characterized by small genomic size? 300 Mbp?and short life cycle.This study focused on BdDREB family.Phylogenetic tree was constructed using the 58 Bd DREB protein sequences and clustered into six major clades,including A1,A2,A3,A4,A5 and A6 subgroups.Chromosomal localization analysis was further performed to systematically investigate the evolutionary features of these BdDREB genes.Additionally,six genes were selected to validate their expressions in different tissues and under different abiotic stress conditions using q-PCR methods,results verified that these BdDREB genes were involved in various developmental and physiological processes.Brachypodium was used as template and encoding full cDNA of DREB gene was cloned?gene ID: XM003570049.3?.The revealed that a 684 bp product was overexpressed in cloned in the plant over-expression vectors;the final vectors were then transformed successively into Arabidopsis.Kanamycin?50mg/ml?was used for resistance screening of transgenic Arabidopsis thaliana seeds grown in ? MS media,furthermore,PCR detection and phenotypic analysis were performed.to confirm the positive transgenic plants.In general,we obtained transgenic Arabidopsis overexpressing BdDREB1 G laying a foundation for further analyses.
