Medicago truncatula,whose genome is relatively small,belongs to the genus leguminosae with high efficiency of genetic transformation.It is also a model legume that completed genome sequencing.Cicer arietinum L is a common leguminous plant in China.The Cicer arietinum L is rich in nutrients,containing a variety of proteins and amino acids.Its genome has been sequenced.The WOX transcription factor is a family of specific transcription factors found only in plants.During plant growth and development,they play an important role.The WOX transcription factors are also involved in abiotic stress.The members of the WOX transcription factor are identified in the whole genome of the Medicago truncatula and Cicer arietinum L by means of informatics.And the phylogenetic analysis of the WOX transcription factor genes of Medicago truncatula and Cicer arietinum L are carried out by software,as well as its protein length,physicochemical properties of the protein,gene structure Analysis,and MEME protein motif analysis,etc.To predict the evolution and function of the family of WOX transcription factor genes of Medicago truncatula and Cicer arietinum L,the function of the family of WOX transcription factors on Cicer arietinum L under salt stress was analyzed by fluorescent quantitative PCR.The results are as follow:By using the Pfam online software and the HMMER online software,the 14 family members were finally screened in the quail genome named as Mt WOX1-Mt WOX14,whose number of amino acids are from 95-590.And 17 family members were finally gotten in the whole genome of Cicer arietinum L named as Car WOX1-Car WOX17,whose number of amino acids are from 184-408.1.Using the software ClustalW2 and MEGA6.0 to carry out the phylogenetic analysis of the family of wolfberry and Cicer arietinum L WOX transcription factor family.The analysis results show that this family of genes can be divided into three categories: the ancient evolutionary branch,the intermediate evolutionary branch,Modern evolutionary branch..2.On-line software GSDS was used to analyze the intron and exon structure of this gene family in Medicago truncatula and Cicer arietinum L.The analysis results showed that the branching structure of the evolutionary tree was highly consistent with the branching structure of Arabidopsis thaliana evolutionary tree.The exon structure of the members of the ancient clade and intermediate clade of the family members is relatively simple and contains only 2-3 exons.The gene structure of modern clade members is more complex and the number of exons is 2-7.The number of introns in the same subfamily of Cicer arietinum L is relatively conservative.There are differences in the number of introns between different subfamilies.And the intron and exonic structure of members in the same clade have certain similarities.From the ancient clade to the modern clade,the CDS region showed a trend of amplification,which shows that the WOX gene family has a diversity of functions during evolution.3.The MEME software was used to analyze the conserved components of this family member.Excluding the Mt WOX10 gene,other members contained both motif1 and motif2 conserved elements.The conserved components of each clade were located in a highly similar position to conserved components.It can be seen that the 17 Cicer arietinum L WOX protein sequences contain the conserved elements motif1 and motif3,and different evolutionary branches contain different structures of the conserved elements.4.Analyzed the expression of six genes of Cicer arietinum L under salt stress by using fluorescent quantitative PCR technology.In result,the expression levels of Car WOX3,Car WOX5,Car WOX7 and Car WOX11 genes were up-regulated in roots.The overall expression levels of Car WOX3,Car WOX4,and Car WOX5 in leaves were low,but Car WOX7's expression was up-regulated,as well as the Car WOX11's expression,at the same time,Car WOX14's expression peaked at 24 h after salt stress.The overall expression level was observed upward during the whole salt stress treatment.
