Caragana intermedia is a kind of mesquite plant that distributes widely in arid and semiarid area,and it has a good ability to resist abiotic stress.Many genes were found playing important roles in stress,growth and metabolism from Caragana intermedia recently.Domian of unkown function(DUF),refer to all the domains which has no known function so far.Among them,DUF220,DUF247 and DUF966 only exist in plants.In this research,three genes that contained DUF from the transcriptome library of Caragana intermedia were selected,and named CiDUF220-1,CiDUF247-1 and Ci DUF966-1respectively.Analyzation with bioinformatics and real-time quantitative PCR had been done.Furthermore,the function of CiDUF966-1 was analysed preliminary,detailed results are as follows:1.Through bioinformatics analysis,we predicted that CiDUF220-1 would not be induced by abiotic stress,Ci DUF247-1 and CiDUF966-1 would be induced by abiotic stress,and the result was verified by real-time PCR.2.CiDUF966-1 locates in the cell membrane.3.CiDUF966-1 overexpression lines compare with wild type and at966-1,the diameter of the rosette is shorter and the rosette curls downwards,the petiole becomes shorter,and the morphology of leaves is round and curly seriously.4.There were not significant differences among wild type,at966-1 and the CiDUF966-1 overexpression lines in resistance to salt stress and drought stress.5.Campared with wild type and at966-1,leaves of CiDUF966-1 overexpression plants showed senescence earlier on 1/2MS mediun,and we ruled out the possibilities of nitrogen deficiency and enthylene senstitivity.
