Based on the previous studies,in order to further understand and obtain the mutation mechanism and molecular information of low energy ion implantation-driven DOB genome,we conducted a comparative genomic study on three low energy ion beam recombinant strains DOB073,DOB113 and DOB981 using bioinformatics methods based on DOB whole genome De novo sequencing.The results showed that the genomes of DOB073 and DOB113 were increased by 12 883 bp and 11 174 bp,respectively,compared with the original strains,but DOB981 was reduced by 223 268 bp and the corresponding genes of the former two were respectively 28 and 23 more than the original strains,but 13 and 18 genes in the genome of the original strain were deleted;while DOB981 reduced 189 genes,there are also new genes.The variant gene analysis and its functional annotations indicated that the functions of novel genes in the low energy ion beam recombination strains DOB073 and DOB113 genomes were mostly related to energy metabolism,environmental adaptation,and DNA damage repair,and are distributed in the anterior,posterior,and middle parts of the genome,respectively.The missing genes are mainly expressed in chromosome separation proteins and chloride channel family proteins,DNA-related metabolism,drug resistance and other related functions,and are enriched in the middle of the genome;for DOB981,the function of the reduced genes is mainly associated with changes in DNA,it is located in the middle of the genome,while rare new genes are associated with DNA metabolism,resistance catalysis,and sulfur metabolism pathways,and are distributed in the anterior and middle parts.The analysis of its phylogeny and evolution showed that the low-energy ion beam recombination strains DOB073 and DOB113 were closely related to the original strains,while the DOB981 genome was fast-growing and had a distant genetic relationship with the original strain.The COG function enrichment analysis of differential genes showed that the unique gene functions of the low-energy ion beam recombinant strains DOB073,DOB113,and DOB981 were significantly enriched in protein translation,ribosome structure and development,cell wall/cell membrane/extracellular layer development,and protective mechanisms etc.in different degree,especially DOB981 is the most special,but carbohydrate transport and metabolism are obviously found in the unique genes of the original strain.This study provided some molecular evidences for the whole-genome mutation and evolution of low-energy ion implantation-driven prokaryotic microorganisms,and also a theoretical and practical basis for the De novo origination of new genes driven by low-energy ion implantation.
