Objective To investigate the single-voxel and multi-voxel magnetic resonance spectroscopy in hippocampus and thalamus of healthy volunteers, and to explore whether the data obtained by the two methods are statistically different. Methods Use the single-voxel and multi-voxel MRS detection and analysis bilateral hippocampus and thalamus of the same age in 50 healthy adult, which N-acetyl aspartic acid(NAA)/creatine(Cr),choline(Cho)/creatine(Cr),NAA/(Cho+Cr)ratio can obtained. Results there were significant differences in Cho/Cr and NAA/(Ch+ Cr)in the left hippocampus between single-voxel and multi-voxel MRS(t=-5.239,4.042,P<0.05);NAA/Cr values were not statistically significant(P>0.05);NAA/Cr and Cho/Cr values in the right hippocampus were significantly different between single-voxel and multi-voxel MRS(t=-3.491,-3.769,P<0.05).There was no statistically significant difference in NAA/ Cho+Cr(P>0.05);The NAA/Cr and Cho/Cr values of single-voxel and multi-voxel MRS were statistically different(t=-2.552,-6.273,P<0.05),and there was no significant difference in NAA/(Cho+Cr)(P>0.05).NAA/Cr,Cho/Cr,and NAA/(Cho+Cr)(t=-5.862,-5.068,-6.316,t=-11.722,-9.077,-9.712,t=-11.314,-9.379,-10.842,P<0.05)were statistically difference in the left thalamus, right thalamus, thalamus with single-voxel and multi-voxel MRS.The NAA/Cr,Cho/Cr,and NAA/(Cho+Cr)levels in the left and right hippocampus of the single-voxel MRS were statistically significally(t=4.369,-5.073,6.959,P<0.05).The NAA/Cr,Cho/Cr,and NAA/(Cho+Cr)levels in the left and right hippocampus of the multi-voxel MRS no statistically significant.(t=1.191,1.503,-0.133,P>0.05).There were no significant difference in NAA/Cr,Cho/Cr,NAA/(Cho+Cr)in Left lateral thalamus with single-voxel MRS(t=1.191,1.503,-0.133,P>0.05),There were no significant difference in NAA/Cr,Cho/Cr,NAA/(Cho+Cr)in Left and right lateral thalamus with multi-voxel MRS(t=-1.141,-0.881,-0.252,P>0.05).There were no significant difference in NAA/Cr,Cho/Cr,NAA/(Cho+Cr)with singlevoxel MRS in normal hippocampus of different genders(t=-1.960,0.122,1.866,P>0.05).There were no significant difference in NAA/Cr,Cho/Cr,NAA/(Cho+Cr)with Multivoxel MRS in normal hippocampus of different genders(t=1.210,-0.493,1.661,P>0.05);there was a statistical difference between Cho/Cr and NAA/(Cho+Cr)in thalamus of different sexes in normal individuals with single-voxel MRS(t=-2.194,2.795,P<0.05),NAA/Cr values were not statistically different(t=1.112,P>0.05);there was a statistical difference between Cho/Cr in thalamus of different sexes in normal individuals(t=-3.27,P<0.05),NAA/Cr and NAA/(Cho+Cr)values were not statistically different with multivoxel MRS(t=-0.580,1.722,P>0.05).Conclusion The single-voxel and multi-voxel MRS are two independent examination methods, and the detection values obtained for the same area of interest are not the same. Therefore, their values cannot be used universally. When the single-voxel MRS was examined, there were inconsistencies between the left and right sides of the normal hippocampus. The study of single-voxel and multi-voxel magnetic resonance spectroscopy on normal human hippocampus is not affected by gender, while the study of the thalamus is influenced by gender.
