The ribosome is an organelle that does not have cell membrane and is the site for protein synthesis.Ribosomes widely exist in various tissues of different organisms,and are consist of rRNAs and ribosomal proteins.rRNAs function as the backbone,while various ribosomal proteins bind to it with non-covalent bond,in which way they're assembled into ribosome large subunits or small subunits.Besides,there are many different kinds of ribosomal proteins.And the homologous ribosomal proteins may possess different functions in different species and in different types of ribosomes.Ribosomal proteins are the important components of the ribosome,the absence of which will affect the function of the ribosome.The loss of important ribosomal protein will lead to the disorder of life activities,the abnormality of biological organs,or even death of the organism.Plant homologous ribosomal proteins are encoded by two or more genes,and the ribosomal proteins encoded by their gene families are functionally redundant or perform different functions.The ribosomal protein L23 also possesses two coding genes,RPL23 AA and RPL23 AB.But so far there is no report about the function differences of the ribosomal protein L23 encoded by these two genes.In this study,Arabidopsis was used as research material to explore the functions of ribosomal protein L23 family.The sequences of the two genes and the proteins encoded by them are found to be extremely similar with comparison of the sequences of RPL23 AA and RPL23 AB genes and their protein products;according to the phenotype of mutant rpl23 aa,mutant rpl23 ab,double mutant RPL23AB/rpl23 ab,RPL23AA/rpl23 aa,it shows that RPL23 AA mutation can result in the malformation of multiple plant organs,while theRPL23 AB mutation has little effect on plant growth,which indicate that the RPL23 AA plays a dominant role in the regulation of plant growth.In addition,if these two genes are mutated at the same time,the vegetative growth and reproductive growth of plants will be seriously affected,indicating that ribosomal protein L23 is important for the normal growth and development of plants;the vetor sepetately constructed with pRPL23AA::RPL23AA,pRPL23AB::RPL23AB and pRPL23AA::RPL23ABcan recover the phenotype of mutant rpl23 aa.It means that the proteins encoded by RPL23 AA and RPL23 AB are functionally redundant,and the promoters of RPL23 AA and RPL23 AB are not responsible for the phenotypic differences between the mutant rpl23 aa and mutant rpl23 ab.GUS staining was performed on 11 day-seedlingsand inflorescences.The vectors were constructed with pRPL23AA::GUS and pRPL23AB::GUS.According to the expression patternsof RPL23 AA and RPL23 AB,these two genes express in various tissues,including roots,stems,leaves and inflorescences,which means ribosomal protein L23 participate in various life activities in different tissues.
