ATP13A3 is one of the P5-ATPase genes which is one of the P-ATPase families' subfamily.Researchers found that there is a protein overexpression in the senescence kidney parenchymal cells.named AFURS1(ATPase family homolog up-regulated in senescence cells).But,we know poor about the mechanism of ATP13A3 in the process of cell senescence.Our experiment use Hela and BJ cell to study the mechanism of ATP13A3 in the process of cell senescence.We identified when knockdown the ATP13A3 protein,p16,p21,and p53 expression is decreased,and increased as the overexpression of ATP13A3 protein.When ATP13A3 is overexpressed,the protein level of ki67 and PCNA is decreased,means the cell's replication is inhibited.In the senescence BJ cell,we knockdown the expression of ATP13A3 protein,find the ?-gal is decreased.We use flow cytometry to detect the uptake of ceramide in the cell when we change the expression of ATP13A3.It indicted that when ATP13A3 protein is overexpressed,the ceramide' fluorescence is decreased.But when we knockdown the ATP13A3 it not increased,about the reason also need the next study.All these date declare that ATP13AA3 protein play a role in the process of cell senescence.
